三疣梭子蟹血细胞ELISA检测试剂盒的研制与应用

摘　　要：为准确、快速检测三疣梭子蟹血细胞数量的变化,利用三疣梭子蟹血细胞单克隆抗体,研制了三疣梭子蟹血细胞间接酶联免疫吸附检测试剂盒。该检测试剂盒最佳反应条件为:血细胞标准样品与单克隆抗体最佳使用比例为血细胞标准样品和单克隆抗体均稀释4倍,试剂盒的蛋白含量检测灵敏度为8μg/mL。应用本试剂盒对健康及人工感染溶藻胶弧菌的三疣梭子蟹血细胞数量进行检测,发现健康蟹子血细胞数量基本不变,人工感染发病蟹子的血细胞数量变化明显。结果证明,该试剂盒能够准确、快速、高效的检测出三疣梭子蟹血细胞数量的变化,并为评估三疣梭子蟹健康状况提供技术手段和理论依据。ELISA kit for rapid diagnosis of portunus trituberculatus haemocyte,it was prepared by employing monoclonal antibody of portunus trituberculatus haemocyte.The best reaction condition of antigen and antibody was that antigen diluted four times,and antibody diluted four times,with a sensitivity of 8μg/mL.The ELISA method was applied to detected quantity change of haemocyte from artificial infected strains portunus trituberculatus and healthy portunus trituberculatus.The quantity of infected was changed significantly,the quantity of health was unchanged.The results indicated that the kit can accurate and efficient to detect the quantity change of portunus trituberculatus haemocyte,and provide promising tool and theoretical basis for the assessment of health conditions of cultured portunus trituberculatus.
