TLR-2和TLR-4受体单核苷酸多态性与鲍曼不动杆菌感染的相关性

摘　　要：目的探讨鲍曼不动杆菌感染与TOLL样受体TLR-2、TLR-4的相关性,为鲍曼不动杆菌感染的早期预防提供依据。方法根据编码序列同义或非同义突变、频率>5%的SNPs位点,以及文献报道的与细菌感染相关SNPs位点作为候选SNPs位点,采用i MLDR技术对423例鲍曼不动杆菌感染病人和385例非感染病人的血标本进行TLR-2、TLR-4受体SNP检测,对各基因位点基因型频数、等位基因频数及单倍型分布情况进行比较和分析。结果所有的SNPs位点符合哈迪-温伯格平衡。6个SNPs位点频率在病例组和对照组间无统计学差异;TLR-4单倍体型GCG与鲍曼不动杆菌感染的相关性有统计学意义(P=0.027);其余TLR2和TLR4单倍体型在实验组和对照组的分布无统计学差异。结论 TLR-4单倍体型GCG与鲍曼不动杆菌感染可能相关。Objective To determine the correlation of SNP polymorphisms of TLR2 and TLR4 gene with Acinetobacter baumanii infection and provide basis for early prevention of Acinetobacter baumanii infection. Methods Four hundred and twenty- three Acinetobacter baumanii infected patients and 385 patients collected from the same ward were selected as expertimental group and control group, respectively. The candidate SNPs at TLR-2（rs3804099, rs7656411 and rs76112010） and TLR-4（rs1927914, rs10759932 and rs11536889） were selected according to their frequency（〉5%）, position, and their associations with infectious diseases reported previously. SNP genotyping was performed using an improved multiplex ligation detection reaction（iMLDR） technique. The relationship between Acinetobacter baumanii infection and SNPs polymorphisms of TLR2 and TLR4 was analyzed by multiple logistic regression model. Results All SNPs were in Hardy-Weinberg equilibrium（HWE）. The hyplotype distribution of TLR2 and TLR4 candidate alleles showed no significant difference between infected patients and controls, while the haplotype GCG of TLR-4 was significantly associated with Ab infection（P=0.027）. Conclusion Our studies suggest that the two SNPs（rs1927914 and rs10759932） of TLR-4 may be related to the susceptibility to Ab infection.
