摘 要:目的研究黄芪对大鼠牙龈β-防御素-2 mRNA表达的影响。方法选取健康6月龄wistar大鼠70只,随机分为7组,每组10只,连续7 d给药,第7 d断颈处死所有大鼠,切取牙龈组织,RT-PCR检测各组大鼠牙龈中β-防御素-2mRNA表达水平。结果 1.78 g/kg组、2.67 g/kg组、3.56 g/kg组、7.12 g/kg组牙龈组织β-防御素-2基因表达上调,且随药物剂量增加而增加;1.78 g/kg组与空白对照组之间,3.56 g/kg组与7.12 g/kg组和阳性对照(脂多糖3 mg/kg)组之间牙龈组织β-防御素-2基因表达水平差异无统计学意义(P>0.05);其余各组间比较差异有统计学意义(P<0.01)。结论中药黄芪对大鼠牙龈β-防御素-2具有诱导表达作用,其诱导表达作用在一定浓度阈内随药物浓度增加而增加。Objective To explore the influence of astragalus on the expression of beta- defensin- 2 in gingiva of wistar rats. Methods 70 healthy 6- month- old wistar rats were divided into 7 groups,each of which included 10 rats. After tail vein injection of astragalus at different dose for 7 days,all the rats were decapitated and the gingiva of rats were collected. The expression levels of Beta- defensin- 2 mRNA in the gingiva were tested by RT- PCR. Results The expression level of beta- defensin- 2 mRNA in gingiva were increased in 1. 78 g / kg treatment group,2. 67 g / kg treatment group,3. 56 g / kg treatment group and 7. 12 g / kg treatment group,which showing a dose dependent manner. Of note,there were no significant difference( P〉0. 05) in the level of beta- defensin- 2 mRNA expression between1. 78 g / kg treatment group and blank control group,as well as 7. 12 g /kg treatment group and positive control( LPS,3 mg / kg) group. Conclusion Astragalus has a function of inducing Beta- defensin- 2 gene expression in gingiva of rat,and the gene expression levels increased with the increase of drug concentration of astragalus in a certain threshold of the drug concentration.
