摘 要:目的探讨黄芪总皂苷(astragalosides,AST)对高糖诱导的大鼠肾小球系膜细胞(MCs)增殖、诱导型一氧化氮合酶(i NOS)表达及一氧化氮(NO)含量的影响。方法将常规培养的MCs分为正常组、甘露醇组、高糖组及高糖+AST(50、100、200μg/m L)组。MTT法观察细胞增殖情况;Western blot法检测各组细胞i NOS表达;Griess比色法检测各组培养上清中NO水平。结果与正常组比较,高糖组培养24、48 h MCs增殖显著,胞内i NOS表达增强(P<0.01)、NO含量增加(P<0.01);与高糖组比较,各浓度AST干预组均能抑制MCs的增殖,并显著抑制i NOS表达、减少NO的含量。结论 AST能抑制高糖诱导下MCs的过度增殖;下调i NOS表达、降低NO含量,从而发挥对糖尿病肾脏的保护作用。Objective To investigate the effect of astragalosides( AST) on proliferation and expression of inducible nitric oxide synthase( i NOS)and nitric oxide( NO) in rat mesangial cells( MCs) cultured with high glucose. Methods Rat mesangial cells were divided into normal group,mannitol group,high glucose group and high glucose +( 50,100,200 μg / m L) AST group. The proliferation of mesangial cells was observed by MTT assay. The expression of i NOS protein was detected by Western blot and the NO level in the supernatant was determined by Griess method. Results Compared with normal group,MCs proliferation and the expression of i NOS and NO increased significantly in high glucose group at 24 h and 48 h( P〈0. 01). The proliferation of MCs and the expression of i NOS and NO were suppressed in high glucose + AST groups compared with HG group. Conclusion AST can suppress the proliferation of MCs cultured in high glucose and decrease the expression of i NOS and NO,which implies AST may protect the diabetic kidney.
