摘 要:目的:探讨龙胆苦苷对雨蛙素诱导的小鼠急性胰腺炎(acute pancreatitis,AP)的治疗作用。方法:采用腹腔连续注射雨蛙素(每次20μg/kg,共5次),建立小鼠AP模型。龙胆苦苷高、低剂量组(300 mg/kg和150 mg/kg)在开始造模后1、5、9 h腹腔注射给予龙胆苦苷,并于小鼠首次腹腔注射雨蛙素后12 h采集样本。观察血清淀粉酶(AMS)和脂肪酶(Lipase)活性,肿瘤坏死因子-α(TNF-α)和白介素-1β(IL-1β)浓度,小鼠胰腺重量系数,组织含水量和胰腺组织形态学的变化。结果:模型组小鼠血清AMS、Lipase活性,TNF-α和IL-1β浓度以及胰腺重量系数和组织含水量均显著升高(P<0.05,P<0.01);龙胆苦苷高、低剂量组腹腔注射给药可显著减弱血清AMS、Lipase活性,降低血清TNF-α和IL-1β浓度,降低急性胰腺炎小鼠胰腺重量系数和组织含水量(P<0.05,P<0.01),改善胰腺组织病理形态学变化。结论:龙胆苦苷具有良好的抗急性胰腺炎作用;其作用机制可能与保护胰腺细胞、抑制炎症介质的激活和释放有关。Objective: To investigate the effect of gentiopicroside on caerulein-induced acute pancreatitis in mice. Methods: Acute pancreatitis (AP) mice model was made by intraperitoneal infusion of caerulein (20 μg/kg) in mice. Gentiopicroside was given by intraperitoneal injection 1,5 and 9 hours after the beginning of intraperitoneal injection of caerulein in mice in both high dose group(300 mg/kg ) and low dose group( 150 mg/kg). Samples were got 12 hours after the first time of intraperitoneal infusion of caerulein in mice. Serum amylase and lipase activity, TNF-α and IL-1β concentrations, the pancreas mass/body mass index, tissue water content and the histopathological changes were determined. Results: Amylase and lipase activity, TNF-α and IL-1 β concentrations, weight coefficient and water content in pancreatic tissue increased significantly( P 〈 0.05, P 〈 0. 01 ) in the model group, but in both high dose and low dose groups, gentiopicroside markedly reduced the pancreatitis-evoked serum amylase and lipase activity, and decreased TNF-α and IL- 1β concentrations, weight coefficient and water content in pancreatic tissue, and improved histopathological changes in pancreatic tissue. Conclusion: Gentiopicroside has the remarkable function of against acute pancreatitis, and the action mechanism may be related to activation and inhibiting the release of inflammatory mediators.
