不同体质MRL/lpr狼疮鼠病情程度及卵巢激素水平的变化

摘　　要：目的:区分MRL/lpr狼疮鼠体质,并观察不同周龄狼疮鼠病情程度及卵巢激素水平变化。方法:将12周龄MRL/lpr小鼠按体质分为寒体组、热体组、常体组,各组分别随机分为A组和B组,每组10只。A组于12周龄时(发病前期)取材;B组于20周龄时(发病高峰期)取材。通过ELISA、放射免疫等检测方法,分别检测24 h尿蛋白、外周血ds-DNA及卵巢激素水平。结果:12周龄时,不同体质MRL/lpr狼疮小鼠24 h蛋白尿程度、外周血抗ds DNA抗体、卵巢孕酮(P)、雌二醇(E2)、睾酮(T)、黄体生成素(LH)、卵泡刺激素(FSH)水平差异无统计学意义(P>0.05),肾脏病理切片HE染色已显示出轻度损害。20周龄时,寒体组24 h蛋白尿程度、外周血抗ds DNA抗体、卵巢E2水平均低于常体组(P<0.05),卵巢T、P、LH、FSH无体质差异(P>0.05),肾脏病理切片HE染色显示出较为明显的肾脏病变,且寒体组肾损害表现较常体组及热体组轻微。与12周龄比较,20周龄常体组及热体组小鼠24 h蛋白尿程度、外周血抗ds DNA抗体水平均明显升高(P<0.01),寒体组小鼠24 h蛋白尿程度、外周血抗ds DNA抗体水平变化无统计学意义(P>0.05),不同体质组卵巢E2、T、LH、FSH均明显升高(P<0.01),但P变化差异无统计学意义(P>0.05)。结论:MRL/lpr狼疮鼠存在明显的体质差异;在发病高峰期,不同体质小鼠的发病程度及卵巢E2水平存在差别。Objective： To distinguish MRL/lpr lupus mice constitution, and to observe the change the severity and ovarian hormones of different week＇ s lupus mice. Methods： 12-week MRL/lpr mice were classified into cold, normal, and hot constitutions, and different constitution group was randomly divided into A group and B group respectively, 10 mice in each group. 12-week old lupus mice （ A group） and 20-week old lupus mice（ B group） was detected. 24 hour urinary protein, blood anti dsDNA antibody and ovarian hormone levels were measured through ELISA, radioimmunnassay detection method. Results： In 12-week old lupus mice, there was no difference between different physical extent of 24 hour urine protein, blood anti dsDNA antibody, P, ovarian E~, T, LH and FSH levels （ P 〉 0.05 ）, but mild impairment appeared on HE. At the age of 20 weeks, in the cold body group the degree of 24-hour proteinuria, blood anti dsDNA antibody, ovarian E2 levels were lighter than normal group （P 〈 0.05）. There were no constitution differences on the levels of ovarian T, FSH and LH. Mice renal biopsies （HE） showed obvious renal lesions and performance of cold body group kidney damage was better than normal group and hot group. 20-week old normal group and hot group mice have more serious 24-hour proteinuria and blood anti dsDNA antibody than the age of 12 weeks （P 〈0.01 ）. Changes of 24-hour proteinuria and the level of anti dsDNA antibody had no statistical significance （ P 〉 0.05 ） in cold body group between different ages. E2, T, LH, FSH were significantly increased in every 20-week group than 12-week group （ P 〈 0.01 ）, but there was no significant difference between the changes of P （P 〉 0.05 ）. Conclusion：There was obvious difference in physical MRL/lpr lupus mice. In the peak of the onset, the illness degree between the different groups and ovarian E2 levels also exist differences.
