摘 要:以蚕豆(Vicia faba)叶片下表皮为材料,结合药理学试验、气孔开度分析和激光扫描共聚焦显微镜技术,探讨磷脂酰肌醇3-磷酸(PI3P)在乙烯诱导蚕豆保卫细胞中H2O2和NO产生以及气孔关闭中的作用。结果显示,磷脂酰肌醇3-激酶抑制剂沃曼青霉素(WM)和LY294002(LY)显著抑制乙烯释放剂乙烯利诱导的蚕豆保卫细胞H2O2和NO产生以及气孔关闭,外源H2O2和NO处理能完全逆转WM和LY对乙烯利诱导的气孔关闭的抑制效应。结果暗示,PI3P通过诱导信号分子H2O2和NO的产生参与了乙烯诱导蚕豆气孔关闭的细胞信号转导过程。By pharmacological analysis, stomatal bioassay and laser-scanning confocal microscopy, the roles of phosphatidylinositol 3-phosphate(PI3P) in ethylene-induced hydrogen peroxide(H2O2) and nitric oxide(NO) production in guard cells and stomatal closure were studied in the epidermal strips of abaxial surface of broad bean(Vicia faba) leaves. The results showed that both wortmannin(WM) and LY294002(LY), inhibitors of phosphatidylinositol 3-kinase, signifi cantly inhibited ethephon(ethylene-releasing compound)-induced production of H2O2 and NO in guard cells and subsequent stomatal closure, while application of exogenous H2O2 and NO completely rescued the inhibitory effect of WM and LY on the ethephon-induced stomatal closure. These results suggest that PI3 P participates in the signal transduction pathway of ethylene-induced stomatal closure via inducing the signaling molecules H2O2 and NO in guard cells of broad bean.
