摘 要:为深入研究沙枣的药用价值,对比沙枣植物基因组DNA提取的方法,选取SDS法、CTAB法、酚—氯仿法以及试剂盒法4种提取方法,通过琼脂糖凝胶电泳对所提取DNA进行完整性检测,通过紫外分光光度法对DNA进行纯度、浓度检测。结果显示,试剂盒法提取DNA的A260/A280值嫩叶为1.83,标本叶为1.81,A260/A230值嫩叶为2.31,标本叶为2.26。研究认为,从纯度与完整度观察分析,试剂盒法为最优方法,经济有效,可在CTAB方法上进行改进。In order to compare methods of extraction genomic DNA from Elaeagnus angustifolia L. , four extraction methods including SDS method, CTAB method, phenol-chloroform method and kit method were selected, and extracted DNA were detected by agarose gel electrophoresis, DNA purity were detected using ultraviolet spectrophotometer. The result showed that the A260/A280 numerical value for young leaves was 1.83, speciens leaves was 1.81, the A260/A230 numerical value for young leaves was 2.31, speciens leaves was 2.26. The kit method was the optimal method, in term of economic and effective, and could be modified in the CTAB method.
