摘 要:目的:观察益气补肾实验方对自然流产模型小鼠母-胎界面和脾脏组织IL-2、IL-4的表达情况。方法:将雌性CBA/J与雄性DBA/2以2∶1合笼交配,建立自然流产模型。将CBA孕鼠随机分为5组:阴性对照组(生理盐水组)、阳性对照组(环孢素组)、中药组(高、中、低剂量中药组)。孕鼠于妊娠第14 d处死,取脾脏和蜕膜、胎盘组织,用ELISA法测母-胎界面及脾脏IL-2、IL-4蛋白含量。结果 (1)在母-胎界面中,与阴性对照组相比,各治疗组IL-2的表达均降低(P<0.05),IL-4的表达均升高(P<0.05);各治疗组中,高剂量组与环孢素组、中剂量组与低剂量组IL-2、IL-4的表达分别比较均无明显差异(P>0.05),高剂量组、环孢素组IL-2的表达较中剂量组、低剂量组低(P<0.05),IL-4的表达较中剂量组、低剂量组高(P<0.05)。(2)在脾脏组织中,与阴性对照组相比,各治疗组IL-2的表达均降低(P<0.05),IL-4的表达均升高(P<0.05);各治疗组IL-2的表达比较均无明显差异(P>0.05),高剂量组、环孢素组、低剂量组IL-4的表达均无明显差异(P>0.05),三者的表达均较中剂量组高(P<0.05)。结论:益气补肾实验方可以上调自然流产模型小鼠母-胎界面与脾脏组织IL-4的表达及下调IL-2的表达,有助于妊娠时免疫耐受状态的维持。Objective: To observe the Chinese medicine with the function of invigorating Qi and tonifying kidney on expressions of interleukin-2 and interleukin-4 at spleen and maternal-fetal interface in abortion-prone mice during pregnancy. Methods: CAB female mice were mated with DBA/2 male mice to establish abortion-prone model,which were randomly divided into five groups: the high dose of Chinese medicine group,the medium dose of Chinese medicine group,the low dose of Chinese medicine group,the negative control group(fed with normal saline),the positive control group(treated with Cyclosprin A). Mice were sacrificed on the 14 th day of gestation. Their spleens,placenta and decidual tissues were taken out to analyze the protein content of interleukin-2 and interleukin-4 by enzyme-linked immunosorbent assay. Results:(1) At the maternal-fetal interface,comparing with the result in negative control group,the expressions of interleukin-2 in all of the therapeutic groups were reduced(P〈0. 05) while the expressions of interleukin-4 in all of the therapeutic groups were increased(P〈0. 05). The expressions of interleukin-2 and interleukin-4 had no obvious difference between the high dose of Chinese medicine group and the positive control group(P〈0. 05),as well as the medium dose of Chinese medicine group and the low dose of Chinese medicine group(P〈0. 05). The expressions of interleukin-2 in the high dose of Chinese medicine group and the positive control group were lower than that of the medium dose of Chinese medicine group and the low dose of Chinese medicine group and the expression of interleukin-4 was the opposite(P〈0. 05).(2) At the spleen tissue,comparing with the result in negative control group,the expressions of interleukin-2 in all of the therapeutic groups were reduced(P〈0. 05) and they had no obvious difference between each other(P〉0. 05).
