摘 要:目的:探讨甘草酸二铵对环磷酰胺诱导小鼠脱毛动物模型治疗的药理作用机制。方法:采用环磷酰胺(Cyclophosphamide,CTX)建立小鼠脱毛模型,甘草酸二铵(Diammonium Glycyrrhizinate,DG)组和生理盐水组脱毛后,分别灌胃DG(100mg/kg)和生理盐水(100mg/kg),持续8周。模型组不作任何处理。运用HE染色、电镜观察脱毛区毛囊的病理形态学改变,利用免疫组织化学法检测小鼠脱毛区CD4+T细胞(CD4+)、CD8+T细胞(CD8+)、INF-γ表达水平。利用流式细胞术检测外周血IL-2、IL-4以及INF-γ表达水平。采用ELISA检测外周血CD4+/CD8+比值。结果:给药8周后,肉眼观察到DG组脱毛区毛发生长与模型组和生理盐水组相比较为浓密;电镜观察到DG组残存毛囊细胞中线粒体模糊、空泡样化程度比模型组和生理盐水组较轻。在各个时间点,DG组脱毛区CD4+、CD8+、INF-γ表达和外周血IL-2、INF-γ表达水平及CD4+/CD8+比值明显低于生理盐水组和模型组(P<0.05),而外周血IL-4表达显著升高(P<0.05)。结论:CTX造成脱毛小鼠T淋巴细胞过度表达,DG通过抑制T淋巴细胞及其分泌的促炎因子过度表达、提高抗炎因子的表达来改善脱毛小鼠毛发的生长情况。Objective: To investigate the pharmacological mechanism of diammonium glycyrrhizinate for alopecia mice model. Methods: Alopecia mouse model was established by using cyclophosphamide,Diammonium glycyrrhizinate(100 mg/kg) and saline(100 mg/kg) were fed separately in DG group and saline group for 8 weeks after alopecia while model group without any treatment. Pathological changes of the hair follicle in alopecia area were observed by using HE staining and electron microscopy. And then CD4+lymphocyte cells,CD8+lymphocyte cells and INF-γ expression levels around the hair follicle in alopecia area were detected by using immunohistochemistry assay. Additionally,IL-2,IL-4 and INF-γ expression levels and CD4+/CD8+ratio were respectively detected by using flow cytometry and ELISA. Results:Compared with the model group and the saline group,the hair of the shed area in DG group was denser after 8 weeks. Electron microscopy confirmed the degree of mitochondrion vague and vacuolar in remaining hair follicle cell in DG group were lighter than model group and normal saline group. At each time point,compared to the saline group and model group,the levels of CD4+,CD8+and INF-γ expressions in shed area and the levels of IL-2 and INF-γ expression and CD4+/CD8+ratio in peripheral blood in DG group were significantly lower(P〈 0. 05) while the levels of IL-4 expression in peripheral blood was significantly higher(P〈 0. 05). Conclusions: T lymphocytes were over-expression in alopecia mice caused by CTX and the condition of hair growth in alopecia mice were improved by inhibiting over-expression of T lymphocytes and its secretion of pro-inflammatory cytokines,increasing the expression of anti-inflammatory cytokines treated with DG.
