摘 要:目的:研究益气活血方对情绪应激情况下心肌缺血模型大鼠血清P物质、ET-1及NO含量的影响,为益气活血药防治心血管疾病提供新的理论设想和依据。方法:采取腹腔注射异丙肾上腺素法[5 mg(kg·d)],慢性不可预见性情绪刺激法制造大鼠情绪应激心血管疾病模型。设空白组、双心疾病模型组、中药组、心肌缺血组、西药组5组。采用超声心电图监测法,记录心脏功能,釆用酶联免疫组化ELISA法测定血清NO、ET-1、P物质的变化。据各组大鼠在实验过程中的情况,分别观察对比用药14 d、32 d后各组相关指标的变化情况。结果:双心疾病模型组大鼠血清NO、ET-1、P物质水平均明显高于空白组(P<0.01或P<0.05);实验32 d后,双心疾病模型组射血分数(EF)和短轴缩短率(FS)均低于空白组,差异有统计学意义(P<0.01);双心疾病模型组、心肌缺血组血清NO、ET-1、P物质含量均高于空白组,差异有统计学意义(P<0.01或P<0.05);中药组、西药组血清NO、ET-1、P物质含量均低于双心疾病模型组,差异有统计学意义(P<0.05)。结论:益气活血药有利于增加情绪应激情况下心肌缺血模型大鼠心脏射血分数,其原因可能与其对P物质、NO和ET-1的作用有关。Objective: To study the effects of Yiqi-huoxue medicine on the lever of serum Substance P (SP),Endothelin 1 (ET-1),Nitric Oxide (NO) in Myocardial Ischemia Model Rats, then to provide a theoretical ideas and basis for Yiqi-huoxue medicine in prevention and treatment of cardiovascular diseases. Method: Adopt the method of intraperitoneal injection of epinephrine [5 mg/(kg·d)], and chronic unpredictability emotional stimuli to make cadiovascular disease rats under emotional stress state. Animals are divided into five groups: blank group, model group, Chinese medicine group, myocardial ischemia group, and western medicine group. Using ultrasonic electrocardiogram, monitoring method to record the heart function, using the quantitative enzyme immunohistochemical-ELISA method to determinate the level of serum NO,ET-land SP. According to these indexes of every group in the experimental process, respectively to observe contrast the changes of different groups after 14 and 32 days, treatment. Results: After 32 days, the ejection fraction (EF) of model group, and fractional shortening (FS) were lower than those of blank group, the difference was statistically significant (P〈0.01); serum NO, ET-1, P contents in model group and myocardial ischemia group were higher than that in blank group, the difference was statistically significant (P〈0.01 or P〈0.05); serum NO,ET-1,P contents in traditional Chinese medicine group and Western medicine group, were lower than the model group, the difference was statistically significant (P〈0.05). Conclusion: The reason why Yiqi-huoxue medicine can increase cardiac ejection fraction in myocardial ischemic rat model under emotional stress condition may be re- lated to its effects on SP,NO and ET-1.
