摘  要:目的研究左归降糖解郁方对糖尿病并发抑郁症大鼠海马星形胶质细胞GFAP及S100B的影响。方法采用高脂乳剂灌胃、尾静脉注射链脲佐菌素(STZ,38mg/kg)、28天慢性应激联合的方法复制糖尿病并发抑郁症大鼠模型,并随机分为5组:模型组,阳性药(二甲双胍0.18g/kg+百忧解1.8mg/kg)组,左归降糖解郁方高、中、低剂量(32.82,16.41,8.20g/kg)组,以正常大鼠为空白组,灌胃给药4周。采用Morris水迷宫测试大鼠空间学习记忆能力,免疫组化法检测海马星形胶质细胞GFAP及S100B的表达情况,Elisa法检测血清中S100B含量。结果与空白组比较,模型组大鼠逃避潜伏期延长,差异有统计学意义(P<0.01);与模型组比较,阳性药组和左归降糖解郁方高剂量组逃避潜伏期缩短,差异有统计学意义(P<0.01)。与空白组比较,GFAP、S100B表达在模型组显著升高,差异有统计学意义(P<0.05);与模型组比较,阳性药组和左归降糖解郁方高剂量组GFAP、S100B表达显著减少(P<0.01或P<0.05)。结论左归降糖解郁方能显著改善大鼠空间学习记忆功能,可能与减少星形胶质细胞反应性增生有关。Objective To investigate the protective effects of salvianic acid A on radiation - induced injury of mice. Methods - BALB/c mice were divided into 5 groups:normal saline control group, radiation saline control,radiation & salvianic acid A 10mg/ kg/day group, radiation & salvianic acid A 20mg/kg/day group and positive control estriol group. Evaluation of the experimental result included the general condition of the mice, survival rate, changes in body weight, RBC, WBC, HGB and PLT counts, spleen weights, spleen index, CFU - spleen. Results 1. Salvianic acid A could protect radiation - induced mice injury and prolong life of the mice obviously. 2. Pretreatment with salvianic acid A significantly increased the RBC, WBC, HGB and PLT counts in irradiated mice. 3. Spleen indexes and CFU - spleen were greatly higher in salvianic acid A pretreated group than others. Conclusion - Salvianic acid A has satisfactory protective effect on radiation - induced mice injury ; the mechanism may be related to the attenuation of hemopoietic system damage.
