摘 要:目的探讨大黄游离蒽醌对重症急性胰腺炎(sever acute pancreatitis,SAP)致大鼠肝损伤的保护作用。方法将雄性SD大鼠54只,随机分为假手术组(A组)、SAP模型组(B组)和SAP模型+大黄游离蒽醌组(C组),采用逆行胰胆管注射3.5%的牛磺胆酸钠制备大鼠SAP模型,于造模前12h及造模后2h以大黄游离蒽醌(20g/kg)2ml灌胃制备给药组,分别于术后6,12,24h处死大鼠后采集大鼠肝组织,HE染色观察肝脏病理组织学改变,ELISA法测定肝组织匀浆中肿瘤坏死因子-α(tumor necrotic factor-a,TNF-α)和白细胞介素-1(interleukin-1,IL-1)水平,髓过氧化物酶法检测肝组织匀浆髓过氧化物酶(myeloperoxidase,MPO)活性。结果与A组相比,B组大鼠肝组织匀浆液的TNF-α、IL-1和MPO水平在三个时间点均显著高于A组(P<0.05);C组大鼠肝组织匀浆液TNF-α、IL-1和MPO水平在6h时显著低于B组(P<0.05),在24h时低于B组,但差异无显著性(P>0.05)。结论大黄游离蒽醌可减轻早期重症急性胰腺炎所致的肝损伤,提示大黄游离蒽醌可能在重症急性胰腺炎所致的肝损伤中发挥着重要作用。Objective To explore the effect of free anthraquinone of rhubarb on rats with sever acute pancreatitis induced hepatic injury. Methods 54 SD rats were randomly divided into three groups,sham operated group( group A),the sever acute pancreatitis group( group B),the sever acute pancreatitis and free anthraquinone of rhubarb group( group C). The sever acute pancreatitis model was induced by retrograde infusion of bilio-pancreatitis duct with 3. 5% sterile sodium taurocholate solution. Free anthraquinone of rhubarb( 20 g / kg) was administrated affusedly down alimentary canal from mouth to stomach with 2ml at 2 hours before and after the inducement of SAP. Collect the liver of every rat in 6h,12 h and 24 h. Observe pathological changes of liver by Haematoxylin and eosin staining. Test the level of TNF-α and IL-1 in liver homogenate by enzyme linked immunosorbent assay. Serum MPO are quantitated by myeloperoxidase method. Results The level of TNF-α,IL-1 and MPO in liver homogenate in group B is significantly increased than group A( P〈0. 05). The level of TNF-α,IL-1 and MPO in liver homogenate in group C is significantly decreased than group B at 6h( P〈0. 05),and the level of TNF-α,IL-1 and MPO in group C is decreased than group B at 24h( P〉0. 05). Conclusion The free anthraquinone of rhubarb can attenuate sever acute pancreatitis induced hepatic injury. The free anthraquinone of rhubarb might play an important role in sever acute pancreatitis induced hepatic injury.
