健脾化瘀方对胃癌MGC-803细胞侵袭转移相关基因表达的影响

摘　　要：目的探讨健脾化瘀方抑制人胃癌MGC-803细胞侵袭转移的作用机制。方法分别采用Real-time q PCR和Western Blot技术检测健脾化瘀方对胃癌MGC-803细胞侵袭转移基因转录和蛋白表达的影响。结果健脾化瘀方作用于人胃癌MGC-803细胞24h后,MMPs、VEGF随着给药浓度增加其mRNA和蛋白表达下降,RECK随着给药浓度增加其mRNA和蛋白表达上升,STAT3基因转录和蛋白表达未见明显变化,而p-STAT3蛋白表达量随着给药浓度增加下降。结论健脾化瘀方具有体外抑制MGC-803细胞侵袭转移的作用,其分子机制可能与抑制STAT3蛋白磷酸化和RECK基因转录、表达,进而影响下游VEGF、MMPs等有关。Objective To study the mechanism of Jiang Pi Hua Yu Decoction in anti-metastasis and anti-recurrence of human gastric cancer cell lines MGC-803. Methods To detect the transcription level of mRNAs and the protein expression of proteins under the treatment of Jian Pi Hua Yu Decoction,basing on the technique of Real-time q PCR and Western blot. Results Jian Pi Hua Yu Decoction down-regulated the expression levels of STAT3,MMPs,VEGF mRNA after 24 h of treatment in gastric carcinoma cell line MGC-803,and the decrease was accompanied by the increased dose. There was a significant difference in each group except the STAT group（ P〈0. 05）,meanwhile,the expression of RECK gene up-regulated by the increased dose（ P〈0.05）. There was no obvious change in the protein expression of STAT in each dose,however,a situation exists that the higher concentration brings the lower expression of p-STAT3,MMPs and VEGF. Furthermore,the RECK one was opposite. Conclusion-Jian Pi Hua Yu Decoction exhibits an obvious potency of anti-metastasis and anti-recurrence in human gastric cancer cell line MGC-803 in vitro. Inhibiting phosphorylation of STAT3,meanwhile increasing transcription level and expression of RECK,to influence downstream MMPs and VEGF may be one of the related molecular mechanism.
