摘　　要：目的 制备单纯疱疹病毒的单克隆抗体。方法 用纯化的单纯疱疹病毒（HSV）抗原免疫雌性BALB/c小鼠,取其脾细胞与骨髓瘤细胞融合。经ELISA间接法筛选和克隆化培养,获得两株分泌抗单纯疱疹病毒单克隆抗体的杂交瘤细胞,纯化其腹水获得单抗,检测该单抗的特异性。结果 这两种单抗与其他相关的病毒抗原无交叉反应,Western blot表明单抗均识别单纯疱疹病毒中相对分子质量50000的蛋白。结论 该单抗为单纯疱疹病毒的单克隆抗体。Objective To prepare the McAb to herpes simplex virus(HSV). Methods Female BALB/c mice were immunized with the purified antigen of Herpes simplex virus, and the spleen cells of them were fused with myloma cells. Two hybridoma cell strains were screened by indirect ELISA and cultured by cloning,and the McAbs secreted by them were purified from the ascites of mice and detected for specificity. Results No cross reaction was observed among the McAbs secreted by the 2 strains and other relevant antigens. Western blot showed that both of them recognized the HSV protein with a relative molecular weight of 50000.Conclusion The preparation is identified as the McAb to HSV.
