青藤碱家兔膝关节皮肤体外释药规律研究

摘　　要：目的对家兔膝关节皮肤进行青藤碱12h体外释放的测定及其释放规律的研究。方法利用体外透皮实验仪模拟体内的环境,以家兔膝关节的皮肤作为渗透媒介,以青藤碱为检测指标,用超高效液相色谱仪测定不同取样点青藤碱的释放量。结果研究表明青藤碱的累计释药量是4.6082(mg·cm-2),累计透过率是58.60%,12h内符合Higuchi方程,方程为Q=1.7357T1/2-1.2501,r值是0.9949。结论青藤碱在家兔膝关节皮肤处12h内释放性良好,家兔膝关节皮肤可用于青藤碱体外释药规律的研究。Objective To determine the 12 h release in vitro of sinomeine and to syudy the rules on the knee skin of rabbits.Methods Using in vitro transdermal environment simulation experimental apparatus the body,Applying knee skin of rabbits as permeability medium and sinomenine as indicator. Determining the different sampling points release rate by ultra performance liquid chromatograph. Results The study showed that the cumulative penetration content of sinomeine was 4. 6082（ mg·cm-2）,the cumulative penetration percentage was 58. 60%. It conformed the Higuchi Equation in 12 h and the equation was Q = 1. 7357T1 /2-1. 2501,r = 0. 9949. Conclusion the releasing of sinomenine on knee skin of rabbits is good in 12 h,the knee skin of rabbits can be use in the release of sinomenine.
