清热化瘀滋阴方对大鼠主动型Heymann肾炎和狼疮小鼠肾组织凋亡蛋白影响

摘　　要：目的:观察清热化瘀滋阴方对大鼠主动型Heymann肾炎(AHN)模型大鼠的肾功能及对狼疮性MRL/Lpr小鼠肾组织Bcl-2、Bax蛋白表达的影响。方法:采用大鼠肾皮质加弗氏完全佐剂腹腔注射建立大鼠主动型Heymann肾炎(AHN)模型,应用清热化瘀滋阴方、泼尼松、清热化瘀滋阴方联合泼尼松,对主动型Heymann肾炎(AHN)模型大鼠进行干预治疗4周,检测其24 h尿蛋白、尿素氮、肌酐;狼疮性MRL/Lpr小鼠给予清热化瘀滋阴方、泼尼松、清热化瘀滋阴方联合泼尼松,治疗4周,检测对肾组织Bcl-2、Bax蛋白的表达。结果:降低大鼠主动型Heymann肾炎(AHN)尿蛋白、尿素氮、肌酐的水平;同时狼疮性MRL/Lpr小鼠肾组织Bcl-2蛋白表达显著降低,显著升高Bax蛋白。结论:清热化瘀滋阴方改善肾功能,促进狼疮肾组织细胞凋亡,可能具有抑制肾组织增生病变作用,发挥保护肾脏的作用。Objective：To investigate the effects of Qingre Huayu Ziyin Prescription on renal function in rats of active heymann nephritis and the expression of Bcl-2 and Bax protein on renal tissue in the MRL/Lpr mice. Methods：AHN rats were produced by intraperitoneal injection of renal cortex and Freund＇s complete adjuvan. Intervention therapy with Qingre Huayu Ziyin Prescription,prednisone,Qingre Huayu Ziyin Prescription＋prednisone were applied respectively to AHN rats for 4 weeks. There after 24 h urine protein,urea nitrogen,creatinine and pathological changes were measured. Intervention therapy was applied to MRL/Lpr mice for 4 weeks in order to detect the expression of Bcl-2 and Bax protein in renal tissue by western blot method. Result：Compare treatment groups with the model group,it was revealed that levels of urine protein,urea nitrogen and creatinine in AHN rats and the expression of Bcl-2 protein on renal tissue in MRL/Lpr mice were significantly decreased. The expression of Bax protein were significantly increased. Conclusion：It has certain therapeutic effect on renal function and can promote lupus renal tissue cell apoptosis. We conjecture it can inhibit hyperplastic lesions on renal tissue and protect kidney.
