摘 要:目的:通过动物体内实验验证加味牛蒡子汤(JWNBZT)对类风湿性关节炎的治疗作用。方法:将20只3月龄左右TNF-Tg小鼠按照体重随机分为模型组和加味牛蒡子汤治疗组,另取10只同窝对照体重相似的野生型小鼠(WT)作为空白对照组,治疗组每天以加味牛蒡子汤灌胃,空白对照组和模型组每天以等量纯净水灌胃,治疗12周后测量小鼠踝关节周长和活动度,并取踝关节制作石蜡切片进行HE染色观察炎性浸润和关节结构改变情况。结果:与WT小鼠相比,TNF-Tg小鼠踝关节明显肿胀(P<0.05),踝关节背伸活动度受限;而加味牛蒡子汤治疗12周明显减轻TNF-Tg小鼠踝关节肿胀(P<0.05)和踝关节活动度(P<0.05)。踝关节石蜡切片HE染色显示空白对照组小鼠踝关节结构完整,没有炎性浸润和骨性侵蚀;模型组小鼠踝关节炎性浸润大量产生,骨侵蚀严重,关节结构明显破坏;加味牛蒡子汤组治疗后炎性浸润和骨结构破坏明显减轻。结论:加味牛蒡子汤能够减轻TNF-Tg关节水肿和结构破坏,是炎症性关节炎治疗的潜在替代药物。Objective: To confirm the efficiency of Jiawei Niubangzi Decoction (JWNBZT) in treating ankle injury of mice with inflammatory arthritis. Method: Twenty TNF-Tg mice at the age about 3 months were ran- domly and evenly divided into model group and treatment group by weight. Ten wild type littermates (WT) withsimilar weight were used as a control group. The treatment group were fed with decoction of JWNBZT once every day, while the other two groups were fed with the same volume of purified water. After 12 weeks' treatment, measured the circumference and the flexibility of the ankle joint. Made paraffin section of the ankle joints of all groups with HE staining to observe inflammatory infihrates and the structural change of the joint. Results: Com- pared with WT littermates, TNF-Tg mice had swollen ( P 〈 0. 05 ) and less flexible ankle joints ( P 〈 0. 05 ), while treated with JWNBZT for 12 weeks, the ankle swelling was alleviated significantly ( P 〈 0. 05 ) and the mobility of the joints was increased ( P 〈 0. 05 ) . Paraffin section with HE staining showed the structure integri- ty of ankle joints without inflammatory infiltrates and bony erosion. Among Mice in the model group, inflamma- tory infiltrates appeared largely and the damage of bone and cartilage in TNF-Tg ankle joints was obvious. Con- chmion: JWNBZT can alleviate inflammation and the damage on the ankle joint structure of TNF-Tg mice. It can be used as a potential alternative drug for inflammatory arthritis.
