摘 要:目的观察并探讨熊胆及熊去氧胆酸对免疫性血小板减少性紫癜(ITP)模型的作用和可能的机制,为熊胆治疗ITP的替代品提供依据。方法利用免疫法建立ITP模型,将小鼠随机分为正常组,模型组,泼尼松组,熊胆组和熊去氧胆酸组,治疗10 d后,测定各组血小板计数,骨髓巨核细胞计数及形态,T细胞亚群,血清细胞因子IL-4、IFN-γ。结果治疗后,熊胆和熊去氧胆酸组均能升高ITP小鼠血小板和产板型巨核细胞计数,升高CD4+/CD8+百分表达比值,降低血清IFN-γ/IL-4比值,与模型组比较差异有统计学意义(P<0.05)。结论熊胆及熊去氧胆酸均能减少血小板及成熟骨髓巨核细胞的免疫性破坏,该作用可能与调节T细胞功能有关;熊去氧胆酸可代替熊胆作为治疗ITP的有效药物。Objective To explore therapeutic effect and its related mechanism of Bear bile and active ingredients UDCA on Immune Thrombocytopenic Purpura(ITP) model, to provide a reference for studying Bear bile and its succedaneum treated on ITP. Methods To establish ITP model by using the method of immunization, all mice was randomly divided into normal group, model group, prednisone group, Bear bile group, UDCA group, after treatment 10 d, observe the platelet number, the changes of the T-cell subgroup, the levels of the Cytokines IL-4 / IFN-γ in each group. Results Bear bile and UDCA can increased the counts of platelet and megakayocyte which can produce platelet, the ratio of CD4^+ / CD8^+, decreased the ratio of IFN-γ / IL-4, compared with the model group, there was particularly significant(P 0.05). Conclusion Bear bile and UDCA can reduce immune destruction of platelet and mature Megakaryocyte, its therapeutic effects may be achieved by regulation of T cellmediated immune.
