摘 要:目的研究AB-8大孔吸附树脂纯化茶树根总黄酮、总皂苷的工艺条件。方法以总黄酮、总皂苷的吸附率和转移率为指标,考察吸附药液质量浓度、药液p H、药材树脂比、洗脱乙醇体积分数、乙醇洗脱用量等影响因素。结果吸附药液质量浓度0.3 g·ml-1,药液p H=6,药材树脂比1:2,依次用12BV水洗,10 BV 30%乙醇洗脱得总黄酮,10 BV 70%乙醇洗脱得总皂苷。结论采用大孔吸附树脂AB-8可较好地纯化茶树根总黄酮与总皂苷,纯化后茶树根总黄酮含量、总皂苷含量均达60%以上。Objective To discuss purification technology conditions of total flavonoids and total saponins from tea root by AB-8 macroporous resin.Methods With absorption rate and metastasis rate of total flavonoids and total saponins as indexes,investigated concentration of sample liquid, p H, ratio of macroporous resin to crude drug, concentration of eluted ethanol and other factors. Results Optimum purification conditions were as follows: Concentration of sample liquid was 0.3 g · ml-1, p H of sample liquid was 6,ratio of crude drug to macroporous resin was1 : 2, total flavonoids and total saponins were eluted by 12 BV water,10 BV 30% ethanol and 10 BV 70% ethanol respectively. Conclusion Macroporous resin AB-8 could be used to purify total flavonoids and total saponins from tea root,the content of total flavonoid and total saponins reached more than 60%.
