Sift6在心肌细胞缺氧／复氧过程中表达的研究

摘　　要：目的通过H9C2心肌细胞系水平缺氧／复氧（H／R）实验，模拟人体心脏灌流水平缺血／再灌注（I／R）过程，探讨Sirtuin6（Sirt6）在心肌细胞H／R过程中的表达变化。方法实验分5组，即正常组（N组），缺氧2h复氧0min（H／R组），缺氧2h复氧30min（H／R30min组）、缺氧2h复氧1h（I-I／R1h组），缺氧2h复氧2h（H／R2h组），Westernblot（WB）法检测Sirt6蛋白表达情况，Real-timePCR检测Sirt6表达最低时Sirtuin家族各亚型mRNA表达情况。结果与N组相比，H／R过程Sift6蛋白表达降低，在复氧1h时最低（P〈0．05）；与N组比较，缺氧2h复氧1h时，Sirt1、2、4、5、6mRNA的表达降低（P〈0．05），其中Sirt6mRNA表达最低（P〈0．05）；Sirt3、7mRNA的表达与N组差异无统计学意义（P〉0．05）。结论Sirt6在H／R过程中表达减少，且呈时间依赖性，在复氧1h时表达最低；．H／R时Sirt1、2、4、5、6mRNA的表达降低，以Sirt6降低最明显。Objective To detect the amounts of Sirtuin 6 （ Sift 6） in H9C2 myocardial cells during ischemia/reperfusion （I/R）. Methods The cells were divided into five groups according to their various treatments ： Group N （without treatment）, Group H/R （2 h of hypoxia without reoxygeration）, Group H/R 30 min （2 h of hypoxia followed by 30 min of reoxygenation）, Group H/R 1 h （2 h of hypoxia followed by 1 h of reoxygenation）, and Group H/R 2h （2 h of hypoxia followed by 2 h of reoxygenation）. The quantities of Sirt6 in each group were measured by Western blotting. The levels of Sirt 1 - 7 mRNA were examined by Real - Time PCR when Sirt 6 expression was at the minimal level. Results Com- pared with Group N, the quantities of Sirt 6 were decreased after H/R, which reached the minimum level after reoxygen- ation for 1 h （P 〈 0.05 ）. Compared with Group N, the levels of Sirt 1/2/4/5/6 mRNA were reduced in Group H/R 1 h （P 〈0.05）, in which the amount of Sirt 6 mRNA was lower than others （P 〈0.05）. However, there was no statistical difference as to Sirt 3/7 mRNA between Group N and Group H/R 1 h （ P 〉 0.05 ）. Conclusion During H/R, the quantity of Sirt 6 is reduced in a time - dependent manner, which reaches the minimum level after reoxygenation for 1 h. The level of Sirt 6 mRNA is lower than that of Sirt 1/2/4/5, through they were all decreased during H/R.
