摘 要:目的研究Bcl-w和Bid蛋白在慢性高原病(CMS)患者骨髓组织中的表达情况,探讨它们在CMS发生、发展过程中的作用。方法选取18例CMS患者为CMS组,应用免疫组织化学SABC法检测骨髓组织中Bcl-w和Bid蛋白的表达情况。16例单纯陈旧性骨折患者为对照组。结果 CMS组骨髓组织中Bcl-w及Bid蛋白阳性表达率分别为77.78%及11.11%,对照组骨髓组织中Bcl-w及Bid蛋白阳性表达率分别为31.25%及50.00%,CMS组骨髓组织中Bcl-w蛋白阳性表达率明显高于对照组(P<0.05);而CMS组骨髓组织中Bid蛋白阳性表达率明显低于对照组(P<0.05)。CMS组Bcl-w、Bid阳性系数分别为(4.15±2.11)及(0.33±0.18),对照组Bcl-w、Bid阳性系数分别为(0.87±0.43)及(0.93±0.51),差异有统计学意义(P<0.05)。CMS组Bcl-w蛋白表达阳性系数与血红蛋白浓度呈正相关(r=0.835,P<0.05),而Bid蛋白阳性系数与血红蛋白浓度呈负相关(r=-0.659,P<0.05)。结论 CMS患者骨髓组织抗凋亡蛋白Bcl-w表达增高,促凋亡蛋白Bid表达降低,Bcl-w和Bid蛋白表达变化与CMS骨髓红细胞凋亡异常有关。【Objective】 To study the expressions of Bcl-w and Bid in bone marrow tissue of patients with chronic mountain sickness(CMS), and explore their possible roles in the developing process of CMS.【Methods】 The samples of bone marrow used in this study were obtained from 18 patients with CMS(CMS group) and 16 patients with old fracture(control group). The expressions of Bcl-w and Bid in the bone marrow tissues were detected using the SP immunohistochemistry technique. The results were analyzed and compared between both groups. 【Results】 The positive rates of Bcl-w and Bid proteins were 77.78% and 11.11%in the CMS group, and 31.25% and 50.00% in the control group, respectively. The expression of Bcl-w in the CMS patients was significantly higher than that in the controls(P〈0.05), but the expression of Bid in the control group was much higher than that in the CMS group(P〈0.05). The positive coefficient of Bcl-w protein in the CMS group was significantly higher than that in the control group [(4.15 ± 2.11) vs(0.87 ± 0.43), P〈0.05]. The positive coefficient of Bid protein in the control group was significantly higher than that in the CMS group [(0.93 ± 0.51) vs(0.33 ± 0.18), P〈0.05]. In the CMS group, hemoglobin(Hb) value was positively correlated with Bcl-w protein and negatively correlated with Bid protein(r = 0.835,-0.659 respectively, both P〈0.05).【Conclusion】 The over-expression of Bcl-w and low-expression of Bid might be involved in mechanism of the change of erythroblast apoptosis in CMS patients.
