摘 要:[目的]比较朝鲜族与汉族丙型肝炎病毒RNA(HCV-RNA)阳性者白细胞介素(IL)28BSNP rs12979860的表型分布,探讨延边朝鲜族易感HCV的可能机制.[方法]选择HCV-RNA阳性的朝鲜族98例、汉族92例及检查证实为肝炎病毒指标阴性的健康朝鲜族53例与汉族50例列入研究对象.应用焦磷酸测序法对IL-28B基因多态性SNP rs12979860位点进行检测,分析朝鲜族与汉族HCV-RNA阳性者及健康人群的C等位基因频率和基因型比例,并对基因型与HCV亚型、HCV-RNA定量、肝功能及血象间的相关性进行分析.[结果]朝鲜族HCV-RNA阳性者中CC型89例、TC+TT型9例,汉族HCV-RNA阳性者中CC型83例、TC+TT型9例;健康人群中CC型92例、TC型11例;朝鲜族与汉族HCV-RNA阳性者及健康组基因频率间无明显差异,基因型与HCV亚型、HCV-RNA定量、肝脏生物化学指标间差异亦均无统计学意义(P>0.05).[结论]延边地区朝鲜族与汉族IL-28BSNP rs12979860位点主要表现为CC型,提示IL-28BSNP rs12979860位点C等位基因频率为优势基因表型,但它不能解释朝鲜族易感HCV的原因.OBJECTIVE To compare to the distribution of IL-28 BSNP rs12979860gene phenotype in patients with hepatitis C virus(HCV)-RNA positive between Korean-Chinese and Chinese nationality of Yanbian area,and to explore the possible mechanism of susceptibility to HCV in Korean-Chinese.METHODSIt was selected as subjective that 98 cases of Korean-Chinese and 92 cases of Chinese nationality with HCV-RNA positive,and 53 cases of Korean-Chinese and 50 cases Chinese nationality with hepatitis virus mark negative as control.The IL-28 BSNP rs12979860polymorphism was detected by pyrosequencing method.The C allele frequency and genotype ratio between the patients with HCV-RNA positive and healthy controls were analyzed,and the correlation among genotype and HCV subtype,HCV-RNA quantity, liver function and hemogram was analyzed.RESULTS There were 89 cases of genotype CC and 9cases of genotype CT+TT in the Korean-Chinese patients with HCV-RNA positive,and 83 cases of genotype CC and 9cases of genotype CT+TT in the Chinese nationality patients with HCV-RNA positive.There were 92 cases of genotype CC and 11 cases of genotype CT in the Korean-Chinese and Chinese nationality healthy people.There were no significant differences between the Chinese nationality and Korean-Chinese patients with HCV-RNA positive in the frequency of C and T allele genotype,and between genotype and HCV subtype,HCV-RNA quantity,liver biochemical index(P〉0.05).CONCLUSIONThe main phenotype of allele frequency of IL-28 BSNP rs12979860gene is CC type in Korean-Chinese and Chinese nationality of Yanbian area,and it indicates that the C allele frequency is the dominant genotype of IL-28Brs12979860 site,but it cannot explain the reason of susceptibility to HCV in Korean-Chinese.
