摘 要:[目的]探讨不同紫外线照射时间对玫烟色棒束孢菌落生长、产孢量、毒力及胞壁疏水蛋白(FAE蛋白)含量的影响,以获得最佳紫外线诱变时间,为深入研究玫烟色棒束孢提供科学依据。[方法]取玫烟色棒束孢sp.1菌株原始菌落配制成1×10^6个/m L的孢子悬浮液恒温培养20 h后,分别取4 m L孢子悬浮液于紫外光灯下照射5、7、10、20、30min,吸取10μL不同照射时间的孢子悬浮液接种于PDA平板培养基培养后测定菌落直径、产孢量、毒力及FAE蛋白含量。[结果]经不同诱变时间处理的sp.1菌株再培养后,其菌落生长和产孢量存在一定差异,其中以诱变10 min处理的孢子生长速度最快、产孢量最多,此诱变时间的菌株对小菜蛾2~3龄幼虫的致死中时间最短,为1.97 d,明显优于其他诱变时间处理的菌株和原始菌株。诱变株FAE蛋白含量也因诱变时间不同有较大差别,其中以诱变10 min处理菌株的FAE蛋白含量最高,达3.46μg/mg,显著高于其他诱变时间处理和对照处理(P〈0.05)。SDS-PAGE分析结果显示,紫外诱变株与对照株之间FAE蛋白条带也存在明显差异,诱变10 min处理在20.0~30.0 k D和66.2 k D处有较高浓度的FAE蛋白。[结论]玫烟色棒束孢具有耐紫外照射的特性,紫外线诱变10 min能使玫烟色棒束孢产生正突变。[Objective ]The effects of UV radiation time on colony growth, spore production, virulence and FAE protein content of/sana fumosorosea were investigated to obtain optimal UV radiation time, in order to provide scientific references for further research on I. fumosorosea. [Method]The 1×10^6 spores/mL spore suspension of L fumosorosea sp. 1 strain was cultivated thermostatically for 20 h, then the 5 cultivated spore suspensions (each 4 mL) were irradiated by UV- light for 5, 7, 10, 20 and 30 min, respectively. And these spore suspensions of each 10 μL were inoculated into PDA mediums, respectively. After culturing, the colony diameter, spore production, virulence and FAE protein content were de- termined. [Result]The results showed that after culturing sp.1 strains irradiated by UV-light for different times, there were some differences in colony growth and spore production to be found, especially, the spores irradiated for 10 min with the highest growth rate and the most quantity showed strong virulence against the 2^rd-3^rd instar P. xylostella larvae for the shortest time (1.97 days), Which was superior to original strain and strains irradiated for 5, 7, 20 and 30 min. In addition, the different radiation times resulted in great differences in FAE proteins contents among different irradiated strains, and the FAE protein content of strains irradiated for 10 min was the highest (3.46 μg/mg), which was significantly higher than FAE protein contents of strains irradiated for 5, 7, 20 and 30 min and control strain. The SDS-PAGE results showed that there were some differences between FAE protein bands of irradiated strains and control strain, and the strains irradiated for 10 min could express high concentrations 20.0-30.0 kD and 66.2 kD FAE proteins. [ Conclusion ]I. fumosorosea is characterized by resistance to UV, which can produce positive mutation after being irradiated by UV-light for 10 min.
