摘 要:本实验旨在探究敖汉细毛羊毛囊的组织结构与形态发生过程,为细毛羊毛囊发育的分子调控机制研究奠定组织学基础。分别采集胎龄第90、120天的胎儿及出生后1 d和30 d的羔羊体侧部(多毛区)和腹股沟部(少毛区)皮肤组织,制作纵、横切片并显微观察。结果表明:敖汉细毛羊毛囊结构包括结缔组织鞘、外根鞘、内根鞘、毛干和毛球部;在胎龄90 d时,体侧部初级毛囊可见皮脂腺原细胞及次级毛囊毛芽,在胎龄120 d时,毛囊形成角质化毛干并穿出体表,再分化次级毛囊发生;出生后1 d和30 d时,穿出体表毛干进一步增多,体侧部初级毛囊密度,在胎龄120 d时低于胎龄90 d时(P<0.01),出生后1 d时低于胎龄120 d时(P<0.05),次级毛囊密度,在胎龄120 d时高于胎龄90 d时(P<0.01),出生后1 d时低于胎龄120 d时(P<0.01),出生后1 d和30 d时差异不显著(P>0.05),S/P比值,在胎龄120 d时高于胎龄90 d时(P<0.01),生后30 d时高于出生后1 d时(P<0.01),对腹股沟部皮肤分析显示,其毛囊密度很低。结果可为了解细毛羊毛囊形态结构变化及筛选与毛密度相关的差异基因提供参考依据。This study aimed to understand the organizational structure of Aohan fine wool sheep hair follicles and hair follicle morphogenesis process, which will lay a histological basis for molecular regulation mechanism of fine-wool sheep hair follicle development. Fetuses at 90 and 120 d of gestation, the lambs at 1 d and 30 d after birth were selected. Then the skin tissues of body side (hairy area) and the groin (Less gross area) were dissected from 12 individuals (3 each stage). The longitudinal and transverse paraffin sections were conducted and then the sections were observed under the microscope. Results showed that the follicle structure of Aohan fine wool sheep consists of connective tissue sheath, outer root sheath, inner root sheath, hair shaft and hair bulb. Primary hair follicle occurred before the gestational age of 90 d. At gestational age of 90 d, the secondary follicle germs and original sebaceous cell could be found. At gestational age of 120 d, hair shaft keratinization and hair shaft through the body surface. Redifferentiation secondary follicles occured at gestational age of 120 d. The primary and secondary follicle hair shafts were further increased at 30 d after birth. Primary hair follicle density of body side at gestational age of 120 d (120 d) was significantly lower than that at gestational age of 90 d(90 d)(P〈0.01), 1 d after birth(0 M ) was significantly lower than that at 120 d (P〈0.05), Secondary hair follicle density of body side at 120 d was significantly higher than that at 90 d(P〈0.01), while at 1 d after birth was significantly lower than that at 120 d (P〈0.01), 0 M and 1 M was not sign- ificantly (P〉0.05); S/P ratio of body side at 120 d was lower that at to 90 d (P〈0.01), 1 M was significantly lower than at 0 M(P〈0.01). Groin skin of fine wool sheep hair follicle density was very low.
