摘  要:本研究旨在分析巴什拜羊DRB3基因遗传多态性与MO感染的相关性。采用PCR-RFLP方法分析巴什拜羊OLA-DRB3基因第2外显子多态性,并计算基因杂合度、多态信息含量和有效等位基因数等遗传参数;通过分析和差异性检验MO阴性和阳性巴什拜羊OLA-DRB3基因型频率和等位基因频率,初步判定MO的抗性或易感性基因。结果表明:巴什拜羊OLA-DRB3第2外显子在第122、154、168和241碱基处表现出多态性;在Taq I、Pst I位点达到Hardy-Weinberg平衡,根据基因杂合度和多态信息含量,表明巴什拜羊具有较为丰富的遗传多样性。在巴什拜羊的24种基因型中,HaeⅢBC(P<0.05)为MO抗性基因,HaeⅢDD(P<0.05)为MO易感基因型;Pst I A(P<0.01)和Pst I B(P<0.01)、HaeⅢC(P<0.01)分别是巴什拜羊MO易感和抗性相关等位基因。此结果可为绵羊抗病育种研究提供理论支持。The aim of the study was to research polymorphisms of the exon 2 of OLA -DRB3 gene in Bashibai Sheep and its association with Mycoplasma ovipneumortiae (MO) infection. The exon 2 gene was amplified by PCR and a uniform fragment of 285 bp was obtained in Bashibai sheep breeds. The PCR product was digested with restriction endonucleases Pst I, Taq I and Hae Ⅲ, respectively. And genetics polymorphism were investigated by PCR-RFLP, compared the genotype differences between the Bashibai sheep with MO infection and that without infection. The results demonstrated extensive polymorphisms of exon 2 of OLA -DRB3 locus at base position 122, 154, 168 and 241 in Bashibai sheep population. The results of x^2 test revealed that Taq I and Pst I restriction endonuclease sites in Bashibai sheep population were fit with Hardy-Weinberg equilibrium. The statistical result of population genetic parameters shows the polymorphism of Bashibai sheep population was high in which the average heterozygosity was 0.541, the average polymorphic information content (PIC) was 0.4854. By comparing alleles and genotypic frequencies of OLA-DRB3 of healthy sheep with that of infectious ones in Bashibai sheep, we found that Pst I B and Hae III C have some resistance to MO(P〈0.01), Pst I A may be susceptible to MO(P〈0.01), genotype Hae Ⅲ BC(P〈 0.05) may be resistant to MO, but Hae Ⅲ DD (P〈0.05)may be susceptible. This paper provides a reference for the disease resistance breeding of sheep.
