摘 要:为全面了解IGF2基因的多态性、系统发育关系以及IGF2蛋白功能位点,本实验利用DNA池结合PCR产物直接测序法和生物信息学对9个鸡品种11种样本的IGF2基因进行系统研究。结果表明:共检测到IGF2基因的9个SNPs,其中3个错义突变导致蛋白性质发生改变;NJ树整体分为三大支,其中绿壳蛋鸡与其他品种遗传差异最大;突变后RNA二级结构、最小自由能和稳定性随之变化;IGF2蛋白存在典型跨膜螺旋区和信号肽,包含2个高度保守的结构功能域IIGF和IGF2-C;同源模建得到了理想可靠的IGF2蛋白三维结构模型。显示,IGF2基因遗传多样性相对较高,突变位点在各群体中分布不平衡,可为进一步研究IGF2蛋白功能奠定理论基础。To understand comprehensively the single nucleotide polymorphism (SNPs) and phylogenetic relationship of IGF2 gene and functional site of the IGF2 protein, IGF2 gene of 11 samples of nine chicken breeds were researched systemly using the DNA pools combined with direct sequencing of PCR products and bioinformatics in the experiment. The results showed that 9 SNPs of IGF2 gene were found, there were 3 missense mutation which led to the nature of protein change. NJ tree was divided into three generally, compared to other varieties , blue-shell chickens' genetic difference was the most biggest one, RNA secondary structure, minimum free energy and structure stability changed consequently after mutating; IGF2 protein had a typical transmembrane helical area and a signal peptide, which included two highly conserved domains IIGF and IGF'2-C, The three-dimensional structure was ideal and reliable got by homology modeling. Conclusion: high genetic diversity was existed in IGF2 gene, mutation sites were unevenly distributed in different samples; the results of the test provided theory basis for further studying the function of IGF2 protein.
