摘 要:为了解鼠伤寒沙门菌L型在猪胆囊内的携带情况,将鼠伤寒沙门菌接种于猪胆汁内进行体外培养,采用非高渗分离培养法分离胆汁培养物内的细菌L型,用二重PCR和核苷酸序列测定检测细菌L型分离物的invA和STM4495基因,建立猪胆囊内鼠伤寒沙门菌L型的检测方法,并对488份猪胆囊标本进行细菌L型的检测。结果表明:在1h至60d的猪胆汁诱导培养物内能够检出细菌L型,其与鼠伤寒沙门菌invA和STM4495基因的核苷酸序列的符合率均为99%;猪胆囊标本中细菌L型的阳性率为51.43%,二重PCR鉴定沙门菌invA基因的阳性率为2.66%,其中鼠伤寒沙门菌STM4495基因的阳性率为1.02%。L-Salmonella Typhimurium was isolated from bile culture inoculated with Salmonella Typhimurium in vitro by non-high osmotic medium, then the detection method of L-Salmonella Typhimurium in pig's gallbladder was established through detecting invA and STM4495 gene of the isolated bile culture by double PCR and nucleotide sequencing and finally 488 pig's gallbladder samples were analyzed by the established detection method to know the carrying condition of L-Salmonella Typhimurium in pig's gallbladder. The results show that L-Salmonella Typhimurium can be detected from pig's bile induction culture inoculated with S. Typhimurium within 1 h~60 d, and the coincidence rate of nucleotide sequence of invA and STM4495 gene between L-Salmonella Typhimurium and S. Typhimurium reaches 99%. The positive rate of L-S. Typhimurium in pig's gallbladder samples is up to 51.43%. The positive rate of invA gene in Salmonella identified by double PCR analysis is 2.66%. The positive rate of STM4495 gene in S. Typhimurium is 1.02 %.
