摘 要:为了研究外源基因转入对动物免疫功能的影响,即转牛Myf6基因小鼠脾脏淋巴细胞的增殖能力以及相关细胞因子IL-2、IFN-γmRNA表达的变化,试验随机选择5只转牛Myf6基因的子一代小鼠,采用MTT法体外检测脾淋巴细胞增殖能力和实时荧光定量PCR研究宿主脾脏中的IL-2mRNA、IFN-γmRNA表达变化。结果表明:Myf6子一代小鼠T淋巴细胞、B淋巴细胞的增殖指数与阴性对照组相比显著降低,IL-2 mRNA相对表达量显著降低,IFN-γmRNA表达也出现了显著下降。说明外源基因Myf6转入抑制了小鼠的脾脏免疫机能。To study the effect of the transfer in of exogenous gene on the immune function in animals, the proliferation ability of spleen lymphocytes and the mRNA expression levels of related cytokines including IL - 2 and IFN - γ in mice transferred with Myf6 genes were investigated. Five mice from the first filial generation of mice transferred Myf6 genes were randomly selected to detect the proliferation ability of the spleen lymphocytes in vitro using MTT method and study the ehanges in the IL - 2 and IFN -γ mRNA expression levels in the host spleen using Real - time fluorescent quantitative PCR. The results showed that the proliferation indexes of T and B lymphocytes in the mice from the first filial generation of mice transferred Myf6 genes were significantly decreased compared with negative control group. The relative expression level of IL -2 mRNA was significantly decreased and IFN -γ mRNAexpression level was also scgnificantly decreased compared with the negative control group. The results indicate that the transfer in of Myf6 as an exogenous gene ean inhibit the immune function of mice.
