摘 要:目的从细胞因子分泌调控水平研究芍药苷(PF)抗炎抗风湿的作用机制。方法收集20例原发性干燥综合征(p SS)患者空腹肘静脉血于肝素抗凝管中,用Ficoll-hypague密度梯度离心分离外周血单个核细胞(PBMC),分为PF干预组和不干预组。体外干预培养,分离细胞培养上清,用酶联免疫吸附试验(ELISA)法检测不同浓度PF对三磷酸腺苷(ATP)刺激下该细胞分泌白细胞介素(IL)-1,IL-6的影响。结果 2、20、80 mol/L PF作用4 h,PBMCs培养上清中IL-1、IL-6水平与不干预组比较,差异均无统计学意义(均>0.05);100、150 mol/L PF作用4 h,PBMCs培养上清中IL-1、IL-6水平与不干预组比较,差异均有统计学意义(均<0.05)。结论 PF可能通过下调p SS患者PBMC的IL-1,IL-6分泌发挥对p SS患者的治疗作用,且具有剂量依赖性。Objective To determine the effects ofpaeoniflorin (PF) on production of ATP-induced pro-inflamma- tory cytokines released by peripheral blood mononuclear cells (PBMCs) in patients with primary Sjgren's syn- drome (pSS). Methods Twenty-two pSS patients were included and their fasting venous blood in the heparin tubes were used to isolate peripheral blood mononuclear cells(PBMC) by FicoU-hypague density gradient centrifugation. There were PF intervention group and non-intervention group. With in vitro culture, cell culture supernatant was in- solated and the amounts ofIL-1β and IL-6 in the supernatant produced by these cells were quantitated by enzyme-lin- ked immuno sorbent assay (ELISA), and then, the regulating effects of PF were evaluated. Results Incubating PBMCs with two, 20, 80μmol/L PF for four hours, there were no statistical significances of IL-1β between PF inter- vention group and non-intervention group[(3 274±90.80)vs(3 405±81.48), (3 214:i:81,12)vs(3 405a:81.48), (3 160±97.63)vs(3 405±81.48), allP 〉 0.05]; Incubating PBMCs with 100, 150 μmol/L PF for four hours, there were statistical significances [(2 819±155.90)vs(3 405±81.48), (2 383±131.6)vs(3 405±81.48), all P 〈 0.05]. Incubating PBMCs with two, 20, 80μmol/L PF for four hours, there were no statistical significances of IL-6 between PF interv ention group and non-intervention group [(55 830±1 680)vs(53 110±1 563), (49 410± 101)vs(53 110±1 101), (49 120± 1 550)vs(53 110±563), all P 〉 0.05]; Incubating PBMCs with 100, 150 μmol/L PF for four hours, there were statistical significances [(46 540d:1 149) vs(53 110±1563), (42 880±1 664)vs(53 110±1 563), allP 〈 0.05]. Conclusions PF has therapeutic effect on pSS patients by reducing secretion of IL-1β and IL-6 in a dose dependent manner.
