摘 要:目的建立胶束电动毛细管色谱法快速检测血浆和尿液中的5种β-内酰胺类抗生素(阿莫西林、头孢氨苄、氯唑西林、青霉素和头孢曲松)含量的方法。方法毛细管为熔融石英毛细管(75/365μm,40/47 cm),运行缓冲液为30 mmol/L SDS的磷酸盐缓冲液(p H=8.0),电压为12 k V,柱温为20℃,检测波长为200 nm。血浆样品经蛋白沉淀和超滤离心等预处理后进样,尿液样品经适当稀释后直接进样分析。结果阿莫西林、头孢氨苄、氯唑西林、青霉素、头孢曲松5种β-内酰胺类抗生素在6 min内完全分离,在浓度为1μg/ml^200μg/ml时,其峰面积和浓度之间的线性关系良好,r分别为0.998 8、0.998 6、0.997 0、0.996 1和0.998 4。尿液和血浆中5种β-内酰胺类抗生素的加标回收率为74%~104%,RSD为2.1%~6.9%。结论该方法具有快速、简便、准确度和精密度高等特点,适用于血浆和尿液中5种β-内酰胺类抗生素含量的快速检测。Objective To develop a new method for the rapid determination of amoxicillin,cefalexin,cloxacillin,benzylpenicillin and ceftriaxone in plasm and urine by micellar electrokinetic chromatography. Methods The capillary was fused silica capillary of 75 /365 μm and effective / total length of 40 /47 cm. The running buffer was 30 mmol / L SDS phosphate buffer at p H8. 0. Separation voltage was 12 k V. Temperature was 20 ℃. Detection wavelength was set at 200 nm. The plasm sample was analyzed after the pretreatment of protein precipitation and ultrafiltration. The urine sample was injected for analysis after proper dilution. Results The amoxicillin,cloxacillin,cephalexin,penicillin,cephalosporin Qu Song of these five β- lactam antibiotics could be completed in 6 min. Good linearity was found within the range of 1 μg / ml- 200 μg / ml,and the r was 0. 998 8,0. 998 6,0. 997 0,0. 996 1 and 0. 998 4 respectively. The recoveries of five β- lactam antibiotics in wistar rat plasm and human urine ranged from 74%- 104%,and the relative standard deviations( RSD) ranged from 2. 1%- 6. 9%. Conclusion This method is fast,simple,precise and it is feasible for the rapid determination of 5 β- lactam antibiotics in plasm and urine samples.
