摘 要:目的评价肉苁蓉颗粒的毒理学安全性。方法采用小鼠急性经口毒性试验、遗传毒性试验(Ames试验、小鼠精子畸形试验、小鼠骨髓微核试验)和大鼠30 d喂养试验,对肉苁蓉的毒理学安全性进行评价。结果以最大给药剂量(20 000 mg/kg体重)进行小鼠急性经口毒性试验,未见小鼠有中毒症状和死亡现象。Ames试验、小鼠精子畸形试验及小鼠骨髓细胞微核试验3项遗传毒性试验结果均为阴性。大鼠30 d喂养试验结果显示,动物总增重、总进食量、食物利用率、血常规和血生化指标与阴性对照组比较,差异均无统计学意义(P>0.05),对大鼠脏器组织均未观察到有害作用。结论肉苁蓉颗粒在本实验条件下未显示有急性毒性、遗传毒性及亚急性毒性作用。Objective To assess the toxicological security of herba cistanches granule. Methods Acute oral toxicity test,mice genetic toxicity tests( Ames test,mice sperm abnormality test,bone marrow cell micronucleus test) and rats 30 days feeding test were conducted to evaluate the toxicological safety of herba cistanches granule. Results No apparent symptom of poisoning and death was observed at the greatest dose of 20 000 mg /( kg·bw) for the acute oral toxicity test. Results of the three genetic toxicity tests( including the Ames test,the mice sperm abnormality test and the bone marrow cell micronucleus test) were negative. There was no significant difference in rat total weight gain,total food intake,food utilization rate,blood count and biochemistry index between treatment groups and negative control group for the 30 d feeding test( P〉0. 05). No apparent pathological injury was observed in rat tissues. Conclusion Herba cistanches granule under the experimental conditions showed no acute toxicity,genetic toxicity and subacute toxicity.
