摘  要:目的了解哮喘对大鼠勃起功能的影响。方法 4周龄健康雄性SD大鼠20只,随机分为哮喘模型组(A)和对照组(B)各10只,实验各组于建模8周后测定阴茎海绵体内压(ICP)、平均颈总动脉压(MAP),测定大鼠血清肿瘤坏死因子α(TNF-α)和血清C反应蛋白(CRP)浓度,截取阴茎海绵体标本检测一氧化氮合酶(NOS)活性、环磷酸鸟苷(c GMP)浓度,并采用Western印迹和实时荧光定量PCR测定阴茎海绵体内皮源性一氧化氮合酶(e NOS)含量和e NOS mRNA的表达情况。结果 A组在临床及组织病理方面均有明显、典型的哮喘表现;ICPmax/MAP×100 A组3V[10.78±8.26]和5V[29.07±6.82]较B组3V[32.75±5.73]和5V[53.71±8.59]均显著降低(P=0.023);血清TNF-!、CRP浓度A组[476±68.45 pg/m L、57.89±27.94 ng/m L]较B组[201.79±67.33 pg/m L、21.09±17.21 ng/m L]显著增加(P<0.001);海绵体NOS活性、c GMP浓度A组[1.17±0.51U/mgprot、36.68±10.29 pmol/mg]较B组[3.64±1.21U/mgprot、53.77±11.08 pmol/mg]显著降低(P=0.034、P=0.041);e NOS mRNA的表达A组[3.98±2.07]较B组[3.07±1.83]差异无统计学意义(P=0.475);e NOS蛋白含量A组(0.42±0.05)较B组(0.81±0.09)显著降低(P=0.039)。结论哮喘可引起e NOS含量和NOS活性下降导致海绵体组织中NO水平和c GMP浓度下降,从而导致大鼠勃起功能障碍。Objective To study the effection of asthma on erectile function in rats. Methods 20 healthy male 4-week-old Sprage-Dawley rats were randomly divided into 2 groups:asthma group( A group) ,control group( B group) . At 8 weeks, all rats underwent erectile function testing by measuring maximum intracavernous pressure /mean arterial pressure ( ICPmax/MAP) ,serum TNF-αand CRP were detected, the NOS activity and cGMP content were examined, eNOS was analyzed by western blot, eNOS mRNA were detected by real-time PCR. Results A group rats represented obvious and typical asthma in clinical and histopathological aspects. The ICPmax/MAP × 100 in group A was significantly less than the level in group B at 3 V [(10. 78 ± 8. 26,and 32. 75 ± 5. 73,respectively)]and at 5V[(29. 07 ± 6. 82,and 53. 71 ± 8. 59,respectively)] stimulation voltage, respec-tively(P=0. 023, P〈0. 001). Serum TNF-aand CRP in group A[(476. 32 ± 68. 45)pg/ml,(57. 89 ± 27. 94)ng/ml] was sig-nificantly increased than these in group B[(201. 79 ± 67. 33)pg/ml,(21. 09 ± 17. 21)ng/ml](P〈0. 001, P=0. 005). NOS ac-tivity and cGMP content in group A[(1. 17 ± 0. 51)U/mg prot,(36. 68 ± 10. 29)pmol/mg] was significantly less than in group B [(3.64 ±1.21)U/ mg prot,(53.77 ±10.08)pmol/mg](P=0.034, P=0.041). The expression of eNOS mRNA in group A [3. 98 ± 2. 07] was no significantly difference with group B[3. 07 ± 1. 83](P=0. 475). The expression of eNOS in group A[0. 42 ±0.05] was significantly less than these in group B[0.81 ±0.09](P=0.039). Conclusion Asthma may mediated erectile dysfunction through the expression of eNOS in corpus cavernosum of rats, simultaneously decrease the activity of eNOS and NOS, and the content of NO and cGMP.
