摘 要:【目的】以新疆主要栽培陆地棉的棉株种子为目标产物,分析不同化学打顶剂处理对棉株种子胚乳萌发的影响。【方法】通过化学打顶剂氟节氨、化学打顶剂金棉科技、未打顶和人工打顶4种不同处理方法,采用蒽酮法、考马斯亮蓝G-250法、蒽酮比色法和印三酮溶液显色法,研究棉株种子胚乳可溶性糖、可溶性蛋白含量、淀粉含量及游离氨基酸含量的变化的生理生化因素。【结果】化学打顶剂氟节胺处理种子胚乳可溶性糖总活性最高,为17%;化学打顶剂金棉科技处理棉株胚乳蛋白质含量最高,其蛋白质含量为1.06 mg/g;喷洒氟节氨化学打顶剂棉株上部种子胚乳含量最高,其淀粉含量为1.89%;人工打顶的棉株种子胚乳游离氨基酸的总活性最高,其游离氨基酸含量为19 333 ug/g。【结论】棉株种子在氟节胺和金棉科技化学打顶剂处理下,与未打顶和人工打顶方法处理棉株种子相比,种子胚乳理化物质积累较多,更有利于棉株种子萌发和代谢活动。【Objective】This experiment mainly aims at cultivating upland cotton seeds in Xinjiang as the target product and analyzing the effects of different chemical detopping treatments on the germination of cotton seed endosperm.【Method】Through the four different treatments: chemical agent in chemical detopping section fluorine ammonia,chemical detopping gold cotton science and technology,non- chemical detopping and artificial detopping,and by anthrone method and Coomassie brilliant blue G- 250 method,anthrone colorimetry,and method of color printing three- ketone solution,the changes of physiological and biochemical factors of the cotton seed endosperm soluble sugar,soluble protein content,starch content and free amino acid content were all researched.【 Result】Experiments found that after section fluorine chemical detopping agent ammonia processed seeds,the endosperm soluble sugar total activity increased by 17%; chemical detopping agent by gold cotton processing increased the endosperm protein content to the highest level,whose protein content was1. 06 mg/g; After spraying section fluorine ammonia chemical detopping agent on the tops of seeds,the endo-sperm content was the highest,and the starch content was 1. 89%; The total free amino acid was the highest by artificial detopping whose free amino acid content was 19333 ug/g. 【Conclusion】The comparison of cotton seeds in ammonia fluoride and in chemical detopping treatment of gold cotton chemical science and technology with seeds of non- chemical detopping treatment,seed physical and chemical endosperm matter accumulated more,which would be more conducive to cotton seed germination and metabolic activity.
