摘  要:【目的】建立高效的新疆早熟陆地棉田间遗传转化与室内潮霉素相结合的筛选体系。【方法】以MS为基本培养基,以去皮早熟陆地棉新陆早17号种子为材料,进行头孢霉素敏感性实验,研究其对棉花植株的影响和抑菌情况;通过在MS培养基上添加不同浓度的潮霉素和IBA,研究新陆早17号最适抗性筛选浓度及最适生根条件。【结果】新陆早17号最适的头孢霉素筛选浓度为500μg/m L,在该浓度下,植株生长不受影响且其鲜重较其他处理增重明显;其对潮霉素反应较为敏感,10μg/m L潮霉素即可抑制棉花根部生长,棉花植株生长矮小叶片发黄;其最适生根培养基为MS+1.0μg/m L IBA,在该浓度条件下生根率为100%,平均根数8.5根;具有3~4片真叶的生根无菌苗经练苗和移栽后成活率为100%。【结论】为新疆早熟陆地棉田间进行"喷花法"的农杆菌遗传转化方式和室内潮霉素抗性筛选鉴定提供了理论基础。【Objective】The study aims to establish the efficient screening system of Xinjiang early maturing upland cotton varieties between genetic transformation in the field and hygromycin selection system indoor.【Method】Cefotaxime sensitivity experiments were done to elucidate the effectiveness of the plant height and bacteriostasis activity by using Xinluzao 17 whose coats were peeled with MS as basal medium,different concentration treatment of hygromycin and IBA were exerted for studying the optimal resistance concentration of hygromycin and the rooting conditions.【Result】The results indicated that the concentration treatment of 500μg /m L cefotaxime had obvious effect on growth and development of Xinluzao 17,and could significantly promote the growth,however strongly inhabitation on the root growth was exhibited with 10 μg/m L hygromycin treatment,and cotton was short and leaves became yellow; The optimal rooting medium was MS medium with1. 0 μg/m L IBA,and rooting rate was 100%,the average root strip- number was 8. 5; The transplantationsurvival rate was 100% after the aseptic seedlings were accommodated to circumstances. 【Conclusion】The implementation of the experiment provided the theoretical foundation for Agrobacterium- mediated genetic transformation of Xinluzao varieties by using the method of " spraying flower" in the field and hygromycin selection system indoor.
