摘  要:[目的]观察端粒酶逆转录酶基因(TERT)慢病度载体对大鼠脊髓损伤的修复及瘢痕形成的影响。[方法]取体重约220 g的清洁级SD大鼠60只,随机分为治疗组、阴性对照组、空白组(单纯损伤)、正常组。采用改良Allen's重物坠落法造模,分别于造模成功后的3 d,1、2、4、6周,5个时间点取大鼠损伤区脊髓,利用免疫印迹(western-blot)法检测GFAP、PCR-ELISA法检测端粒酶的表达及免疫组化技术观察胶质瘢痕的形成。[结果]不同治疗方式其BBB评分结果存在差异,有统计学意义(P<0.01),且各对照组与干扰组差异均有统计学意义(P=0.000)。干扰组大鼠损伤脊髓GFAP表达低于其余各组,且逐渐减少,阴性及空白对照则增多;端粒酶的表达在干扰组中降低,标记NF-200荧光的,TERT基因慢病毒载体干扰组及正常组其荧光密度明显高于阴性对照组及空白组。[结论]作用于大鼠脊髓损伤区的TERT慢病毒载体对脊髓损伤的修复有促进作用,可以减少损伤区胶质瘢痕的形成,为脊髓损伤的治疗研究提供进一步实验依据。[ Objective] To observe the effect of telomerase reverse transcriptase gene (TERT) lentiviral vectors for spinal cord injury repair and scar formation. [ Method ] Sixty clean SD rats with the weight about 220g were randomly divided into treatment group, the negative control group, the control group (simple injury), the normal group. The Modified Allen's weight drop method was adopted to make model. Western - blot was used to detect the GFAP. PCR - ELISA was used to detect the te- lomerase expression and immunohistochemical observation of glial scar on at 3 d and 1,2,4 ,and 6 weeks after operation. [ Re- suit ] There were significantly statistical differences among Bundle branch block (BBB) scores of different treatments (P 〈 0. 01 ). Moreover,there was statistical significance between each the control groups and combined interference group (P = 0.000). The GFAP expression was lower in interference group than that in other groups. In the negative and blank control groups, it in- creased. Telomerase expression reduced in interference group. As to NF - 200 fluorescence marker, interference group and the normal group was significantly higher than that of its fluorescence intensity of negative control group and the control group. [ Conclusion] TERT lentiviral vectors seems to promote repair of spinal cord injury and can reduce the formation of glial scar in the damage zone. It provides further experimental evidence for the research in treatment of spinal cord injury.
