摘 要:目的建立HPLC法测定烈香杜鹃不同药用部位中4种黄酮类成分的含有量。方法烈香杜鹃甲醇提取液的分析采用Kromasil C18色谱柱(4.0 mm×250 mm,5.0μm);流动相为甲醇-0.1%磷酸水溶液(50∶50);体积流量1.0m L/min;检测波长360 nm。结果烈香杜鹃花、叶和嫩枝中的金丝桃苷、槲皮素、山柰酚、异鼠李素分别在0.061 2~1.224μg(r=0.999 8)、0.052 6~1.052μg(r=0.999 9)、0.010 5~0.210μg(r=0.999 6)和0.020 6~0.412μg(r=0.999 4)范围内具有良好的线性关系,平均回收率分别为98.5%(RSD为2.3%)、98.6%(RSD为2.8%)、95.7%(RSD为1.3)、97.1%(RSD为2.7%)。结论该方法准确、灵敏,而且烈香杜鹃嫩枝中不仅槲皮素的含有量较高,同时也存在山柰酚和异鼠李素,具有一定的药用价值,能为其临床应用提供理论依据。AIM To establish a method for determining four flavones in different medical parts of Rhododendron anthopogonoides by HPLC. METHODS The analysis of R. Anthopogonoides methanolic extract was carried out on Kromasil C18column( 250 mm × 4. 6 mm,5 μm) with methanol-0. 1% phosphate aqueous( 50 ∶ 50) as mobile phase,the flow rate was 1. 0 m L / min,and the detection wavelength was set at 360 nm. RESULTS The linear ranges of hyperoside,quercetin,kaempferol and isorhamnetin existed in flowers,leaves and twigs of R. Anthopogonoides were 0. 061 2- 1. 224 μg( r = 0. 999 8),0. 052 6- 1. 052 μg( r = 0. 999 9),0. 010 5- 0. 210μg( r = 0. 999 6) and 0. 020 6- 0. 412 μg( r = 0. 999 4),respectively. Their average recoveries were 98. 5%( RSD was 2. 3%),98. 6%( RSD was 2. 8%),95. 7%( RSD was 1. 3%) and 97. 1%( RSD was 2. 7%),respectively. CONCLUSION This method is accurate and sensitive,and the twigs of R. anthopogonoides have high content of quercetin along with kaempferol and isorhamnetin,having medicinal value. So it can provide theoretical basis for clinical application.
