UPLC法测定地黄叶及块根中的梓醇和毛蕊花糖苷

摘　　要：目的比较地黄叶及块根中梓醇和毛蕊花糖苷的含有量。方法地黄叶及块根甲醇提取液的分析采用ACQUITY UPLC BEH Shield RP18色谱柱(2.1 mm×100 mm,1.7μm);柱温35℃;流动相为乙腈-0.1%磷酸水溶液,梯度洗脱;体积流量0.2 m L/min;检测波长210 nm。结果梓醇含有量由高到低,依次为地黄嫩叶(149.22 mg/g)>老叶(49.29 mg/g)>块根(33.33 mg/g)>干叶(15.53 mg/g);毛蕊花糖苷含有量依次为地黄老叶(10.27 mg/g)>干叶(10.03 mg/g)>嫩叶(9.56 mg/g)>块根(0.28 mg/g)。结论地黄嫩叶中梓醇的含有量约为老叶的3倍,但毛蕊花糖苷的含有量略低于老叶。地黄叶存放一年后,毛蕊花糖苷的减少量比梓醇小,表明前者稳定性高于后者。而且,这两种成分在叶中的含有量均比在块根中更高。AIM To compare the contents of catalpol and verbascoside in leaves and root tubers of Rehmannia glutinosa Libosch. METHODS The analyses of methanolic extract of R. Glutinosa leaves and root tubers were applied on ACQUITY UPLC BEH Shield RP18column（ 2. 1 mm × 100 mm,1. 7 μm） with acetonitrile-0. 1% phosphoric acid aqueous as mobile phase in a gradient elution mode,column temperature was maintained at 35 ℃,flow rate was 0. 2 m L / min,and detection wavelength was set at 210 nm. RESULTS Catalpol contents were in sequence of R. glutinosa tender leaves（ 149. 22 mg / g） 〉matured leaves（ 49. 29 mg / g）〉 root tubers（ 33. 33 mg / g） 〉dried leaves（ 15. 53 mg / g）,while verbascoside contents were in sequence of R. glutinosa matured leaves（ 10. 27 mg /g）〉 dried leaves（ 10. 03 mg /g） 〉tender leaves（ 9. 56 mg /g） 〉root tubers（ 0. 28 mg /g）. CONCLUSION The catalpol content in tender leaves of R. Glutinosa is about three times as many as that in matured leaves,but the verbascoside content is slightly lower than that in the latter. In addition,after leaves stored for one year,the decrease in verbascoside content is slower than that in catalpol content,showing that the stability of verbascoside is higher than catalpol. At the same time,the contents of catalpol and verbascoside are higher in leaves than those in root tubers.
