摘 要:目的明确单次热应激对小鼠睾丸增殖细胞的影响。方法 6周龄雄性C57小鼠36只,随机分为热处理组和对照组。小鼠热处理组单次43℃水浴,干预15 min,于1、7和14 d取材,HE染色观察睾丸结构形态,免疫组织化学染色检测增殖细胞定位及变化,Western Blot检测Ki67蛋白表达,并进行统计学分析。结果应激处理组生精小管排列疏松,曲细精管中细胞数量明显减少甚至消失,细胞散落在曲细精管中;免疫组化可见,增殖细胞呈现动态变化过程,单次热应激处理后1 d增殖细胞显著减少,第7、14天增殖细胞数量开始慢慢恢复;Western Blot可见KI-67表达量在热应激处理后7 d表达量显著降低,第14天表达量慢慢恢复,且各组之间差异有统计学意义(P<0.05)。结论小鼠睾丸经43℃水浴单次热应激15 min后,增殖细胞的动态恢复是后期精子生成的结构基础。Objective To investigate the effects of heat shock treatment on the survial of germ cell in mice. Methods Six C57 male mice were randomly divided into control group and heat shock- treated group. The heat shock- treated group was submerged for 15 min in a water bath set at 43 ℃,and collected samples at 1 d,7 d and 14 d. The testes from each animal were collected make HE stained slices. The expression of KI67 in testis was detected by immunohistochemistry. Western Blot to detect protein expression and to make a statistical analysis. Results HE staining showed that one day after heat shock treatment,the structures of spermatogenic epithelium were disordered. The germ cell in seminiferous tubules was incompact. Seven days after heat shock treatment,seminiferous tubules atrophied and loosely arranged. The germ cells and the sperm in the tubules were significantly reduced or even disappeared. Fourteen days after heat shock treatment,seminiferous tubules were arranged more closely,and the number of germ cells increased. Western Blot detection had the same phenomenon,and there had significant difference in two groups(P〈0. 05). Conclusion Mice testes by 43℃ water bath single thermal stress after 15 min,germ cells dynamic recovery are base of the sperm production.
