摘  要:目的利用基因芯片技术研究三氧化二砷(As2O3)对小鼠大脑神经免疫相关基因表达水平的影响。方法将小鼠按体质量随机分为3组,其中两组进行自然饮用含As2O3自来水方式染毒:4mg/L As2O3染毒组为高剂量组,1mg/L As2O3染毒组为低剂量组,另一组为生理盐水对照组。小鼠染毒60d后,对其大脑组织进行总RNA抽提、纯化、体外转录合成cRNA探针、cRNA探针生物素标记、与Mouse Genome 430 2.0Araay基因芯片杂交,扫描杂交信号,最后进行芯片数据处理和生物学信息分析。结果与对照组比较,染砷组小鼠大脑组织白细胞介素家族基因有43%下调,其中,高剂量染砷组小鼠大脑组织的IL11、IL20、IL25和IL1a的表达比对照组分别下调18.38、19.20、24.25和8.00倍。与对照组比较,下丘脑-垂体-肾上腺轴受体相关基因Crhr1和Crhr2、AVP及Avpr1b、Avpr1a、Avpr1a、Avpr2的表达均明显下调。结论亚慢性砷暴露可能明显下调小鼠大脑组织白细胞介素家族部分基因和下丘脑-垂体-肾上腺轴受体相关基因的表达,今后应使用更多动物样本并从蛋白水平研究这个问题。Objective To study the effects of As2 O3 on expression level of genes related to neuroimmunization in brain of mice by genechip.Methods The rats were randomly divided into three groups:4mg/L arsenic trioxide(As2O3),1mg/L As2O3,and control groups.After 60 days,expressions of target genes in cerebrum of the mice were studied by genechip.Results Compared to the control group,43% of the interleukin family genes in brain of the mice exposed to arsenic were statistically down-regulated.IL11,IL20,IL25 and IL1ain the brain tissue of the mice exposed to high dose arsenic were down-regulated by 18.38,19.20,24.25 and 8.00 times respectively compared to the control group.Compared to the control group,the expression of hypothalamic-pituitary-adrenal axis receptor related genes Crhr1 and Crhr2,AVP and Avpr1 b,Avpr1a,Avpr1 a,Avpr2were significantly down-regulated.Conclusions The gene expressions of interleukin family and hypothalamus-pituitary-adrenal axis receptor can be down-regulated in the mouse subchronicly exposed to arsenic.It is necessary to verify the differential expression in protein level on more mice.
