摘 要:目的探讨对氨基水杨酸钠(PAS-Na)对染锰大鼠血清脂质过氧化、血和骨骼金属元素水平的影响。方法28只SD雄性大鼠被随机分为对照组、染锰组、低、高剂量PAS-Na治疗(L-、H-PAS)组。染锰组、L、H-PAS组腹腔注射(ip)Mn Cl2·4H2O 15 mg/kg,对照组ip等容量生理盐水,每日1次,每周5 d,连续3周。然后,L、H-PAS组背部皮下注射(sc)PAS-Na 100和200 mg/kg,对照组和染锰组背部sc等容量生理盐水,每日1次,连续5周。测定血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性、丙二醛(MDA)含量,测定血、股骨、髂骨锰(Mn)、铁(Fe)、锌(Zn)、铜(Cu)、钙(Ca)和镁(Mg)含量。结果与对照组比较,染锰组血清MDA含量增高,H-PAS组MDA含量降低(P<0.05)。血、股骨、髂骨Mn含量比对照组高,L-PAS组比对照组低(P<0.05);染锰组血Fe、Mg含量均高于对照组,Zn含量低于对照组,L-PAS组Mg含量降低(P<0.05);染锰组髂骨Cu、Mg含量均低于对照组(P<0.05)。结论过量锰暴露可致机体产生脂质过氧化,干扰Fe、Cu、Mg、Zn含量的分布。PAS-Na有促排锰作用,对锰致血脂质过氧化和镁含量影响有干预作用。Objective To explore the effects of sodium para-aminosalicylic acid( PAS-Na) on lipid peroxidation in serum and metal elements in blood and bone of manganese( Mn)-exposed rats. Methods 48 SPF healthy male Sprague-Dawley rats were randomly divided into normal control group,Mn-exposed group low and high dose,PAS-Na treatment( L- and H-PAS) group. Mn-exposed group,L-PAS and H-PAS group received intraperitoneal injection( ip) of Mn Cl2·4H2O 15 mg / kg,while the control group ip physiological saline in the same volume,once a day,all at 5 days a week for 3 weeks. Then,L-PAS and H-PAS group rats received back subcutaneous injection( sc) of PAS-Na 100 and 200 mg / kg respectively,while the rats in the Mn-exposed group and the control group received back sc of physiological saline at the same volume once a day,for 5 weeks. The activity of antioxidant enzymes( SOD,GSH-Px) and content of malonaldehyde( MDA) in serum and Mn,Fe,Zn,Cu,Ca and Mg of blood,femur and iliac bone were detected. Results Serum MDA content of Mnexposed group increased when compared with the control group,but H-PAS group decreased the serum MDA content induced by Mn(P〈0. 05). Mn level in blood,femur and iliac bone of Mn-exposed group were higher than those of the control group,the treatment in the L-PAS group decreased Mn in blood,femur and iliac bone induced by Mn(P〈0. 05). Compare with the control group,Fe and Mg levels in blood of the Mn-exposed group increased significantly,while Zn in blood decreased(P〈0. 05). And Mg levels in blood of L-PAS decreased significantly when compared with the Mn-exposed group(P〈0. 05). Cu and Mg levels in iliac bone of Mn-exposed group decreased obviously when compared with those of the control group(P〈0. 05). Conclusion Mn exposure can lead to lipid peroxidation,and disturb the distribution of Fe,Zn,Cu,Ca and Mg. PAS-Na can effectively remove Mn and inhibit lipid peroxidation and Mg content induced by Mn.
