摘 要:CD154分子是肿瘤坏死因子(TNF)家族中的一个成员,它与其受体CD40结合后介导产生一系列免疫效应,在体液免疫及细胞免疫应答中发挥着重要作用。CD154分子表达水平与其介导炎症应答强度密切相关。同TNF家族成员一样,CD154分子的表达调控主要集中在转录水平,受诸多转录因子的直接调控。这些转录因子通过调控CD154表达水平,在CD154介导相关疾病中发挥重要作用。CD154分子异常表达及其诱导的炎症反应是包括自身免疫系统疾病、心血管疾病、糖尿病和肿瘤等多种疾病在内的重要发病机制之一。在转录层面对CD154分子表达进行调控将是临床治疗上述疾病的一个关键干预手段。CD154 is a member of tumor necrosis factor (TNF) supergene family. The CD40-CD154 engagement plays a vital role in both humoral and cellular immunity. The intensity of the CD154-induced inflammatory responses was tightly associated with the expression level of CD154 molecules. Regulation the expression of CD154 was mainly at the level of transcription, which was similar to other TNF members, and it was tightly controlled by a series of transcriptional factors (TFs). Recently, aberrant CD154 expression and the CD154-in- duced inflammatory responses have been noted in a series of diseases including autoimmune disorders, diabetes, atherosclerosis and cancer. Aberant CD154 expression and the related in flammation were also involved in pathogenesis of these diseases. Therefore transcriptional regulation of CD154 will be a beneficial therapy in these CD154-related disorders.
