仙附温阳通络饮对Buerger Disease大鼠红细胞表面CR1含量及CD44、CD59百分率的影响

摘　　要：目的观察仙附温阳通络饮对血栓闭塞性脉管炎(Buerger Disease)大鼠红细胞表面补体受体1(CR1)含量和CD44、CD59百分率的影响,并初步探讨其免疫作用机制。方法选用SPF级雄性Wistar大鼠72只,平均随机分成假手术组、模型组、仙附温阳通络饮低、中、高剂量组及脉络宁颗粒组。股动脉注射月桂酸钠溶液造模,次日开始灌胃给药,各组药物均由蒸馏水配制,模型组和假手术组只给服蒸馏水。15 d后取材,采用酶联免疫吸附法检测血清中CR1含量变化、流式细胞术检测新鲜外周血红细胞表面CD44、CD59百分率变化。结果模型组血清中CR1含量及外周血红细胞表面CD44、CD59百分率显著降低,与假手术组比较,差异有统计学意义(P<0.01);仙附温阳通络饮中剂量组治疗后CR1含量及CD44、CD59百分率显著升高,与模型组、脉络宁颗粒组比较,差异有统计学意义(P<0.01)。结论仙附温阳通络饮可有效调控血栓闭塞性脉管炎模型大鼠红细胞表面CR1含量及CD44、CD59百分率,其作用机制有别于脉络宁颗粒,与增强红细胞免疫功能有关。Objective To observe the impacts of xianfu wenyang tongluo decoction on the content of complement receptor 1（ CR1） of and the percentages of CD44 and CD59 on erythrocyte membrane in the rats of thromboangiitis obliterans（ Buerger＇ s disease） and preliminarily explore the mechanism on immunization.Methods Seventy- two SPF male Wistar rats were selected and equally randomized into a sham- operation group,a model group,the low- dose group,the middle- dose group and high- dose group of xianfu wenyang tongluo decoction,and a mailuonin granules group. The femoral artery injection with sodium silicate solution was used for modeling. On the next day,the intragastric administration was applied. The medicines were prepared with distilled water in each group. Only distilled water was used in the model group and the sham- operation group. Fifteen days later,the materials were collected. ELISA was used to detect CR1 content in serum and flow cytometry was to detect the percentages of CD44 and CD59 on erythrocyte surface in peripheral blood. Results In the model group,serum CR1 content and CD44 and CD50 percentages on erythrocyte membrane in peripheral blood were reduced significantly,presenting the significant differences as compared with the sham- operation group（ P〈 0. 01）. In the middle- dose group,after treated with xianfu wenyang tongluo decoction,CR1 content and CD44 and CD59 percentages were increased significantly,presenting the significant differences as compared with the model group and the mailuoning granules group（ P 〈0. 01）.Conclusion Xianju wenyang tongluo decoction effectively regulates CR1 content on erythrocyte membrane and the percentages of CD44 and CD59,and its effect mechanism is related to the enhancement of erythrocyte immune function,different from that of mailuoning granules.
