摘  要:目的探讨粉防己碱(Tet)对雌激素受体(ER)阴性人乳腺癌MDA-MB-435S细胞的作用。方法 MTT方法检测Tet对MDA-MB-435S细胞体外生长的影响。用碘化丙啶单染流式细胞术测定法检测Tet对MDA-MB-435S细胞周期的影响。结果 Tet对MDA-MB-435S细胞生长的抑制作用随着Tet浓度的增加、时间的延长而增大,Tet剂量与时间之间存在交互作用(P<0.01)。Tet作用于MDA-MB-435S细胞24 h后,G0/G1期细胞比例均显著增多,随Tet浓度增加,G0/G1期细胞比例增多;S期、G2M期细胞比例均明显降低,并且随Tet浓度增加,S期、G2M期细胞比例降低。结论 Tet对雌激素受体阴性乳腺癌细胞系MDA-MB-435S增殖有明显抑制作用,且存在浓度和时间依赖性。Tet阻滞MDA-MB-435S细胞于G0/G1期,降低S期比例,抑制DNA合成是其抑制乳腺癌细胞生长的机制之一。Objective To discuss the effect of tetrandrine(Tet)on MDA - MB - 435S cell of ER negative human breast cancer. Methods MTT was used to determine the impact of Tet on the in vitro growth of MDA - MB - 435S. The propidium iodide simple staining flow cytometry was used to determine the impact of Tet on the cell cycle of MDA - MB - 435S. Results Regarding the inhibitory effect of Tet on the growth of MDA - MB - 435S,the effect was getting stronger while Tet concentration was increasing and the time was prolonging. The interaction happens between Tet dose and time(P ﹤ 0. 01). In 24 h when Tet acting on MDA- MB - 435S,the cell proportion at G0 / G1 was increased significantly. With the increasing of Tet concentra-tion,the cell proportion at G0 / G1 was increased. The cell proportion was decreased at S stage and G2 M stage and was getting decrease when Tet concentration was increased. Conclusion Tet inhibits apparently the pro-liferation of MDA - MB - 435S in ER negative breast cancer cell and presents the concentration - time de-pendence. Tet blocks MDA - MB - 435S at G0 / G1 and reduces the proportion at S stage. The inhibition of DNA synthesis is one of the mechanisms on the inhibition of breast cancer cell growth.
