摘 要:目的观察固本止咳中药对COPD模型小鼠肺组织中角质细胞生长因子(Keratinocyte Growth Factor,KGF)表达的影响,探讨其治疗COPD的作用机制。方法采用鼻腔滴入LPS加熏香烟的方法建立COPD小鼠模型,实验分为空白对照组、模型对照组、固本止咳中药组;运用免疫组化方法检测小鼠肺组织中KGF表达水平,ELISA方法检测小鼠肺组织匀浆上清液中KGF的分泌水平。结果肺组织中KGF表达检测显示,模型对照组较空白对照组明显减少(P<0.05),经固本止咳中药治疗后,固本止咳中药组较模型对照组明显升高(P<0.05)。结论固本止咳中药促进COPD模型小鼠肺组织黏膜损伤的修复作用的机制之一是通过促进KGF的分泌实现的。Objective To observe the effect and mechanism of Gubenzhike Recipe on KGF expression in lung of COPD mice. Methods The COPD mice model was established by exposure to cigarette smoke in combination with intra - nostrils injection of lipopolysaccharide. The mice were divided into three groups : negative control group, model group and Gubenzhike Recipe group. Use immunohistochemistry to test the expression of KGF in lung tissue of COPD model. Use the ELISA to test KGF level in supematant of lung homogenate. Results The expression of KGF in lung tissue decreased in model group ( P 〈 0.05 ). The expression of KGF in lung tissue was higher in Gubenzhike Recipe group than those in model group ( P 〈 0.05 ). Conclusion One of the mechanisms of improving lung tissue mueosal repair after injury of Gubenzhike Recipe is achieved through promoting the secretion of KGF.
