摘 要:[目的]利用ISSR分子标记构建盾叶薯蓣品种鉴定的DNA指纹图谱,为盾叶薯蓣药材鉴定和良种选育提供技术支持。[方法]筛选ISSR引物,对10个盾叶薯蓣品种进行PCR扩增和电泳检测,统计条带并进行遗传多样性分析、聚类分析等;选择核心引物,建立DNA指纹图谱鉴定体系。[结果]从50条ISSR引物中筛选出9条,对10个品种扩增共得到86条谱带,多态性条带72条,多态性条带比率为83.72%;选出3条ISSR核心引物建立了10个薯蓣品种的ISSR指纹鉴定图谱。[结论]盾叶薯蓣品种遗传多样性丰富;3条核心引物所构建的植物图谱中,每个品种均有各自特异的共有谱带、特有谱带、缺失谱带,这些谱带的组合可用于盾叶薯蓣品种鉴定。[Objective]With the aim to identify Dioscorea zingiberensis varieties,ISSR molecular markers was used to get its DNA fingerprint.[Method]ISSR primers were screened out,the PCR and polyacrylamide gel electrophoresis were carried out for the 10 D. zingiberensis. varieties,genetic diversity analyses and cluster analyses were performed,and the core primers were selected to set up an identified fingerprint system. [Result]9 primers with clear band and high polymorphism were selected from 50. A total of 86 bands were amplified from 10 D. zingiberensis varieties,72 bands among them were polymorphic,the percentage of polymorphism band( PPB) was 83. 72%. Three core primers were select out to set up fingerprint of 10 D. zingiberensis Varieties. [Conclusion] The genetic diversity of D. zingiberens varieties was abundant. Each of 10 variety has its universal bands and specific bands,these unique bands can be used in the identification of the D. zingiberensis varieties.
