摘 要:目的探讨血管内皮生长因子(VEGF)和p53蛋白(p53)表达与子痫前期(PE)发病及其严重程度的相关性。方法选取2013年2月-2014年12月在该院产科行剖宫产的产妇90例,采用酶联免疫吸附测定法(ELISA)检测产妇血清中VEGF和p53的表达,应用反转录聚合酶链反应(RT-PCR)检测胎盘中VEGF和p53基因的表达。其中对照组(正常妊娠产妇)30例,轻度子痫组30例,重度子痫组30例。结果同对照组相比,轻度子痫组和重度子痫组患者血清中VEGF和p53表达降低,差异有统计学意义(P〈0.05);与轻度子痫组相比,重度子痫组患者血清中VEGF及p53表达降低,差异有统计学意义(P〈0.05)。不同组别血清中VEGF和p53表达呈正相关(r=0.319,P〈0.01)。同对照组相比,轻度子痫组和重度子痫组患者VEGF和p53基因表达降低,差异有统计学意义(P〈0.01);同轻度子痫组患者相比,重度子痫组患者血清中VEGF和p53基因表达降低,差异有统计学意义(P〈0.05)。不同组别中VEGF和p53基因表达呈正相关(r=0.179,P〈0.05)。结论PE患者血清中VEGF和p53的表达水平有不同程度降低,可作为该病的检测指标。发病机制可能与VEGF和p53基因表达水平降低有关。Objective To explore the correlation between the expressions of vascular endothelial growth factor( VEGF) and p53 protein and onset and severity of preeclampsia( PE). Methods From February 2013 to December 2014,a total of 90 puerpera receiving cesarean section in this hospital were collected. Enzyme linked immunosorbent assay( ELISA) was performed to determine serum VEGF and p53 levels in these women. Reverse transcription polymerase chain reaction( RT- PCR) was used to detect gene expressions of VEGF and p53. The women were divided into control group( normal puerpera,30 cases),mild preeclampsia group( 30 cases),severe preeclampsia group( 30 cases). Results Compared with control group,the expression levels of VEGF and p53 in serum of mild preeclampsia group and severe preeclampsia group decreased,and the difference had statistical significance( P 〈0. 05); compared with mild preeclampsia group,the expression levels of VEGF and p53 in serum of severe preeclampsia group decreased,and the difference had statistical significance( P〈 0. 05). The protein expression levels of VEGF and p53 in different groups were positively correlated with each other( r = 0. 319,P〈 0. 01). Compared with control group,gene expression levels of VEGF and p53 in mild preeclampsia group and severe preeclampsia group decreased,and the difference had statistical significance( P〈 0. 01); compared with mild preeclampsia group,gene expression levels of VEGF and p53 in severe preeclampsia group decreased,and the difference had statistical significance( P 〈0. 05). Gene expression levels of VEGF and p53 in different groups had positive correlation with each other( r = 0. 179,P 〈0. 05). Conclusion The expression levels of VEGF and p53 in PE patients decreased in different degrees,which can be used as an indicator for this disease. The pathogenesis of PE may be related to reduction of VEGF and p53 gene expressions.
