摘 要:目的通过中等强度跑台运动干预,探讨运动经ERβ和RANKL/OPG对去卵巢大鼠骨量的影响效应,为阐明有氧运动对骨量的影响及机制提供实验依据。方法 3月龄雌性SD大鼠随机分为BC(基础)、SHAM(假手术)组、OVX(去卵巢)组、OVX+T(去卵巢+运动)组,后3组各分为4、8、12周组。OVX+T组采用跑台训练,坡度5°,5d/周。以DXA测定大鼠全身BMD;以RT-PCR测定ERβmRNA表达;以Western blot测定ERβ、RANKL和OPG蛋白表达。结果运动组大鼠BMD随运动时间增长而增加,OVX+T12组BMD显著高于同期OVX组(P<0.01)。OVX+T各组大鼠ERβmRNA表达量均显著高于同期SHAM组,OVX+T8组达到最大值,且显著高于同期OVX8(P<0.05)。SHAM各组大鼠ERβ、OPG蛋白表达量均显著高于同期OVX组(P<0.01);RANKL蛋白表达量均显著低于同期OVX组(P<0.01)。OVX+T各组大鼠ERβ、OPG蛋白表达量表现为上升趋势,均为OVX+T12组达到最大值;RANKL表达量呈现下降态势,OVX+T12组达到最低值。结论中等强度跑台运动上调骨ERβ、OPG蛋白及ERβmRNA表达,下调RANKL蛋白表达,降低RANKL/OPG,增加去卵巢大鼠BMD;运动增加去卵巢大鼠骨量的机制与ERβ表达增加和RANKL/OPG比值下降密切相关。Objective To determine the effects of moderate intensity treadmill running on bone mass by ERβ and RANKL/ OPG in OVX rats, and provide experimental basis for exploring themeehanismof midentisenty aerobic training on bone mass. Methods 3 -month -old female SD rats were divided into groups at random, included basalcontrol (BC) group, SHAM4, 8, 12 (pseudooperation 4, 8, and 12 weeks), OVX4, 8, 12(Ovarieetomy 4, 8, and 12 weeks), OVX +T4, 8, 12 (Ovari- ectomy + training 4, 8, and 12 weeks). OVX + T groups respectively received 4, 8, and 12 weeks aerobic training on tread- mill (5 d/week, slope 5). BMD was measured by dual energy X- ray absorptiometry(DXA). ERβ mRNA of femur was detected by RT - PCR. RANKL and OPG level of femur were detected by western blot. All data were analyzed using SPSS 16.0 statistical software. Results BMD showed growing along with the training time in OVX + T. BMD in OVX + T12 showed significant higher than that in OVX12 (P 〈 0.01 ). BMD in OVX + T8 and OVX + T12 showed no marked difference than that in SHAM8 and SHAM12. Compared resepera - tedly with SHAM4, SHAM8 and SHAM12, ERβ mRNA was signifi- cantly eleveted in OVX + T4, OVX + T8 and OVX + T12 respectively (P 〈 0.05 ), the maximum value appeared in OVX + TS, which highered remarkedly than that in OVX8. ERβ and OPG were signifi - cantly eleveted in each group of SHAM (P 〈0.01 ), RANKL was lowered than that in same time OVX group (P 〈 0.01 ). ERI3 and OPG expression showed rising trend in eachgroup of OVX + T, the maximum value appeared in OVX + T12, which was no significance than that in SHAM12; RANKL expression showed downtrend in each group of OVX + T, and the minimum value appeared in OVX + TI2, which was no significance than that in SHAM12. Conclusions The moderate intensity treadmill running intervention up - regulated ERβ mRNA, ERβ and OPG expression, down - regulated RANKL expression in femur of OVX rats, and re- duced RANKL,/OPG.
