摘  要:目的观察1型糖尿病胃轻瘫大鼠模型建立过程中相关指标的变化。方法 3月龄雄性wistar清洁级大鼠30只,采用数字表法随机分为模型组(2%链脲佐菌素30mg/kg,多次腹腔注射),对照组(腹腔注射等量的0.9%氯化钠溶液)。美蓝灌胃实验测定胃排空率,每周观察并记录体重、24h饮水量、食量、尿量及血糖。结果与对照组比较,造模后,模型组大鼠各个时点的平均空腹血糖浓度均明显升高(均P<0.01),模型组大鼠的胃内色素残留率明显高于对照组(P<0.01),并伴有饮水量、尿量增多,食量减少,体重减轻,以及食量、精神、活动、皮毛、大便性状的改变。结论 2%链脲佐菌素30mg/kg剂量多次腹腔注射建立T1DM大鼠模型,并通过不规则高脂饮食诱导糖尿病胃轻瘫(diabetic gastroparesis,DGP)模型方法是可行的,为今后基于DGP模型的研究提供了一定的数据支持。Objective To establish rat type I diabetic gastroparesis (DGP) model and to observe the relative indexes. Methods Thirty Wistar rats were randomly divided into model group and control group. DGP model was induced by intraperitoneal injection of streptozotocin (STZ) 30mg/kg and same volume of saline was injected in rats in control group. Gastric emptying rate was determined by methylene blue gavage experiment. The body weight, 24-h drinking volume, urine volume and food intake were recorded every week. Results Compared with normal group, the blood sugar of model group was significantly higher (P〈 0.01), the gastric emptying rate was significantly lower in model group (P〈0.01). In model group the water intake and urine volume increased, food intake and body weight decreased, associated with the changes of vigor, fur appearance and stool features. Conclusion Type 1 diabetic DGP model has been successfully established by injection of STZ and high-fat irregular diets.
