摘  要:目的研究羊蹄总皂苷(BJS)对小鼠肝损伤的保护作用及其机制。方法将健康小鼠随机分为空白对照组、模型组、联苯双酯组、BJS低中高剂量组(100、200、400 mg/kg),连续给药7 d;末次给药2 h后,腹腔注射0.1%的CCl4橄榄油溶液10 ml/kg,建立小鼠急性肝损伤模型;观察BJS对急性肝损伤小鼠肝脏指数、组织病理学改变的影响;采用ELISA法检测小鼠血清中丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)以及肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)、白介素6(IL-6)和肿瘤坏死因子-α(TNF-α)含量的变化。结果与模型组相比,BJS治疗组肝脏指数明显降低,肝组织病理学改变明显改善;BJS能显著降低血清ALT和AST含量(P<0.05,P<0.01),降低肝组织中MDA、IL-6及TNF-α含量(P<0.05,P<0.01),上调GSH和SOD活性(P<0.05,P<0.01)。结论 BJS对CCl4引起的小鼠肝脏损伤有保护作用,其保护机制与减少自由基产生,提高抗氧活酶活性以及降低炎症细胞因子TNF-α和IL-6有关。Objective To study the protective effect of rumex japonicus total saponins(RJTS)on liver injury in mice and its mechanism. Methods The healthy mice were randomized into a blank control group,a model group,a bifendate group,and RJTS low - dose,middle - dose and high - dose groups(100 mg/ kg,200 mg/ kg,400 mg/ kg). The medicine was given successively for 7 days. Two hours after the last medication,0. 1% CCl4 olive oil solution 10ml/ kg was injected intraperitoneally to establish the model of a-cute liver injury in mice. The impacts of RJTS on liver indexes and histopathological changes were observed in the mice of acute liver injury. ELISA was used to determine the levels of ALT and AST in serum,SOD,MDA, GSH,IL - 6 and TNF - α in liver tissue. Results Compared with the model group,in RJTS treatment groups,liver indexes were reduced apparently and the histopathological changes of liver were improved obvi-ously. RJTS reduced significantly serum ALT and AST levels(P ﹤ 0. 05,P ﹤ 0. 01),reduced the levels of MDA,GSH,IL - 6 and TNF - α in liver tissue(P ﹤ 0. 05,P ﹤ 0. 01)and up - regulated GSH and SOD activi-ty(P ﹤ 0. 05,P ﹤ 0. 01). Conclusion RJTS presents the protective effect on liver injury in mice and its pro-tective mechanism is relevant with reducing free radical production,improving antioxidant enzyme activity and reducing inflammatory cell factors,TNF - α and IL - 6.
